2型糖尿病相关LncRNA及其靶蛋白在外周血中的差异表达及诊断价值研究
Differential expression of type 2 diabetes mellitus-associated LncRNAs in peripheral blood and their diagnostic value
【摘要】 目的:通过检测2型糖尿病(Type 2 Diabetes Mellitus,T2DM)患者外周血中LncRNA n385775、LncRNA n341954、LncRNA n336302、LncRNA n408376的表达水平及靶蛋白ALOX5AP、BCL2A1蛋白表达水平,分析其与各指标的相关性,探讨目标LncRNA的诊断价值。方法:(1)收集2023年10月—2024年3月就诊于石河子大学第一附属医院的T2DM患者及健康人群的全血及血浆,通过RT-qPCR检测LncRNA的表达水平及ELISA检测靶蛋白ALOX5AP、BCL2A1蛋白表达水平;(2)采用Spearman相关性分析探讨目标LncRNA与各指标之间的相关性;(3)采用单因素及多因素Logistic回归分析T2DM的影响因素;(4)采用ROC曲线评价目标LncRNA在T2DM中的诊断价值。结果:(1)T2DM组LncRNA n385775、LncRNA n341954的表达水平高于对照组,LncRNA n336302、LncRNA n408376 2组间比较差异无统计学意义;(2)LncRNA n385775的表达水平与结局指标、靶蛋白ALOX5AP呈正相关,与HOMA-β呈负相关;LncRNA n341954的表达水平与结局指标、HOMA-IR、靶蛋白BCL2A1呈正相关,与HOMA-β呈负相关;(3)多因素Logistic回归显示LncRNA n385775、LncRNA n341954升高是T2DM的危险因素;(4)ROC曲线显示LncRNA n385775诊断T2DM的AUC为0.728,调整后AUC为0.901;LncRNA n341954的AUC为0.793,调整后AUC为0.871。结论:LncRNA n385775、LncRNA n341954在T2DM患者中的表达量升高,可作为诊断T2DM的生物标志物。
【Abstract】 Objective: This study aim to investigate the expression levels of four long noncoding RNAs(LncRNAs;n385775, n341954, n336302, n408376 in peripheral blood of patients with type 2 diabetes mellitus(T2DM), as well as the expression levels of their target proteins ALOX5 AP and BCL2A1. We further analysed their correlation with clinical indicators and explore the diagnostic value of the target LncRNAs. Methods: (1)Whole blood and plasma were collected from T2DM patients and healthy individuals who attended the First Affiliated Hospital of Shida from October 2023 to March 2024. The expression levels of LncRNAs were detected by RT-qPCR, and the protein levels of ALOX5 AP and BCL2A1 were determined by ELISA.(2)Spearman’s correlation analysis was used to evaluate the relationships between target LncRNAs and clinical indicators.(3) The influencing factors of T2DM were analyzed by univariate and multivariate logistic regression.(4)The diagnostic value of the target LncRNAs in T2DM was evaluated using ROC curve analysis. Results:(1)The expression levels of LncRNA n385775 and LncRNA n341954 in the T2DM group were higher than those in the control group, while no significant differences were observed for LncRNA n336302 and LncRNA n408376.(2)The expression level of LncRNA n385775 was positively correlated with clinical indicators and target protein ALOX5 AP, and negatively correlated with HOMA-β; the expression level of LncRNA n341954 was positively correlated with clinical indicators, HOMA-IR, and target protein BCL2A1, and negatively correlated with HOMA-β.(3)Multivariate logistic regression showed that increased expression of LncRNA n385775 and LncRNA n341954 were risk factors for T2DM.(4)ROC curve analysis showed that the area under the curve(AUC) of LncRNA n385775 for diagnosing T2DM was 0.728, and the adjusted AUC was 0.901. The AUC of LncRNA n341954 was 0.793, and the adjusted AUC was 0.871.Conclusions:Elevated expression of LncRNA n385775 and LncRNA n341954 in patients with T2DM may serve as biomarkers for the diagnosis of T2DM.
