咬合干扰致小鼠口颌面痛敏及抑郁障碍的研究
Effects of occlusal interference on both orofacial pain and the depression-like psychological states of mice
【摘要】 目的:建立一种新的咬合干扰模型,探讨小鼠咬合干扰对不同性别小鼠口颌面疼痛及其抑郁心理的影响。方法:我们采用垫片粘接的方式将厚度约1 mm的塑料垫片粘接于C57BL/6J小鼠的左下最后一颗磨牙上。通过测量小鼠的体重变化观察小鼠的适应性。通过用0.04 g的von Frey细丝刺激小鼠面颊部,我们记录下小鼠出现躲避或攻击等阳性反应的次数以反映小鼠的口颌面痛敏情况。强迫游泳实验(forced swimming test, FST)用来观察咬合干扰对小鼠抑郁心理的影响。结果:在高强度咬合干扰建立初期,小鼠体重有下降趋势后逐渐平稳并回升。咬合干扰模型建立后3 d小鼠阳性反应次数较造模前出现明显差异(P<0.05)。小鼠的颌面部机械痛行为的显著性差异从第3 d持续到第14 d。雄性小鼠和雌性小鼠口颌面部机械痛敏行为无明显差异。与对照组小鼠相比,口颌面疼痛模型组雄性小鼠和雌性小鼠在强迫游泳实验中漂浮不动时间明显增加。对照小鼠则活动时间更长。而雄性小鼠组和雌性小鼠组的抑郁心理行为无明显差异。结论:咬合干扰的确会造成小鼠的口颌面部疼痛及抑郁心理的产生,雄性小鼠与雌性小鼠的口颌面痛敏及抑郁心理行为无明显差异。
【Abstract】 Objective:To establish a new occlusal interference model and explore the effects of occlusal interference on both orofacial pain and the depression-like psychological states of male and female mice.Methods:We bonded a plastic shim with a thickness of approximately 1 mm to the left lower last molar of C57BL/6J mice using a bonding method.The adaptability of the mice was observed by measuring their body weight changes.The mechanical pain behavior of the orofacial region of the mice was tested by 0.04 g filaments,and the number of positive responses(avoidance and attack) of the mice was recorded.Through the forced swimming test(FST),we observed the impact of occlusal interference on the depression-like psychological states of the mice.Results:In the initial stage of establishing high-intensity occlusal interference,the body weight of the mice showed a downward trend,and then gradually stabilized and rebounded.After the establishment of the occlusal interference model,the number of positive responses showed a significant difference compared with that before the model was established from the 3rd day after the operation(P<0.05).The significant difference in the mechanical pain behavior of the orofacial region of the mice lasted from the 3rd day to the 14th day.There is no significant difference in the trends of mechanical pain behavior changes between male and female mice.Compared with the control group of mice,in the first two minutes after entering the water,both male or female mice in the orofacial pain model group had a significantly increased floating immobility time in the forced swimming test.Control mice had a longer active time.There was no significant difference in depression-like psychological behaviors between the male and female mice groups.Conclusions:Occlusal interference can indeed cause orofacial pain and depression-like psychological states in mice.There is no significant difference in the mechanical hypersensitivity and depression-like behaviors between male and female mice.
