核因子E2相关因子2介导的氧化应激在腹膜透析相关性腹膜纤维化中的研究进展
Progress of nuclear factor E2-related factor 2 mediated oxidative stress in peritoneal dialysis-associated peritoneal fibrosis
【摘要】 腹膜纤维化(PF)是腹膜透析患者超滤衰竭的重要原因,氧化应激(OS)在PF中发挥重要作用。核因子E2相关因子2(Nrf2)作为维持细胞氧化还原稳态的关键转录因子,参与调节了腹膜间皮细胞转分化、新生血管生成、炎症反应等多种过程。文章主要围绕Nrf2通过调节OS调控PF的具体机制,如抑制OS介导的EMT、抑制OS相关的炎症反应、抑制OS介导的新生血管生成、抑制OS相关线粒体稳态失衡以及靶向Nrf2的潜力与挑战等方面进行综述。
【Abstract】 Peritoneal fibrosis(PF) is an important cause of ultrafiltration failure in peritoneal dialysis patients, and oxidative stress(OS) plays an important role in PF. As a key transcription factor for maintaining cellular redox homeostasis, nuclear factor E2-related factor2(Nrf2) is involved in the regulation of peritoneal mesenchymal transition(EMT), angiogenesis, inflammation and other processes. This article mainly reviews the specific mechanisms by which Nrf2 regulates PF by regulating OS, such as inhibition of OSmediated EMT, inhibition of OS-related inflammation, inhibition of OS-mediated angiogenesis, inhibition of OS-related mitochondrial homeostasis imbalance, and the potential and challenges of targeting Nrf2.
