ZNF750在宫颈鳞状细胞癌中的表达特征研究
Study on the expression characteristics of zinc finger protein 750 in cervical squamous cell carcinoma
【摘要】 目的 探究锌指蛋白750(zinc finger protein 750,ZNF750)在宫颈鳞状细胞癌(cervical squamous cell carcinoma, CESC)中的表达特征。方法 在mRNA水平,使用癌症基因组图谱(The Cancer Genome Atlas, TCGA)的307例CESC的RNA-seq数据,分析ZNF750在CESC组织中的表达水平,并使用单细胞RNA测序(single-cell RNA sequencing, scRNA-seq)和空间转录组学技术分析ZNF750在不同细胞类型中的表达情况。在蛋白水平,采用免疫组化法检测ZNF750在62对CESC和非CESC对照的表达差异。通过DEPMAP数据库获取规律间隔成簇短回文重复序列(clustered regularly interspaced short palindromic repeats, CRISPR)敲除数据,研究ZNF750基因对CESC细胞生长的影响。结果 RNA-seq中ZNF750 mRNA在CESC组织中的相对表达值为(5.2015±2.1754),高于非癌对照组织[(1.3219±1.5677),P=0.01;曲线下面积(AUC)=0.912];单细胞测序和空间转录组分析结果也显示CESC细胞中ZNF750呈高表达。免疫组化实验发现ZNF750蛋白在CESC组织[表达中位数为12.0(11.2,12.0)]表达高于非CESC组织[4.0(2.8,4.4),P<2.22×10-16,ROC AUC→1.00]。细胞增殖方面,敲低ZNF750可显著抑制CESC细胞系(SIHA、C4II、HCS2等)的增殖(CRISPER score<0)。结论 ZNF750的表达上调可能在CESC的发生发展中发挥促癌作用。
【Abstract】 Objective To explore the expression characteristics of zinc finger protein 750(ZNF750) in cervical squamous cell carcinoma(CESC).Methods At the mRNA level,307 cases of RNA-seq data of CESC from The Cancer Genome Atlas(TCGA) were analyzed to determine ZNF750 expression in CESC tissues,additionally,single-cell RNA sequencing(scRNA-seq)and spatial transcriptomics were used to characterize the expression pattern of ZNF750 across diverse cell types.At the protein level,immunohistochemistry was performed to detect the different expression of ZNF750 in 62 pairs of CESC tissues and non-CESC controls.Using clustered regularly interspaced short palindromic repeats(CRISPR)knockout data retrieved from the DEPMAP database,we investigated the impact of the ZNF750gene on the proliferation of CESC cells.Results The relative expression value of ZNF750 mRNA in CESC tissue was(5.2015±2.1754),which was higher than that in non cancer controls based on RNA-seq data mining [(1.3219±1.5677),P=0.01,AUC=0.912].Single-cell level analysis and spatial transcriptomics results showed that in CESC tissues,ZNF750 expression levels were highly expressed in CESC cells.Immunohistochemistry revealed that the median expression level of ZNF750 protein in CESC tissues was 12.0(11.2,12.0),which was markedly higher than that in non-CESC tissues [4.0(2.8,4.4),P <2.22 × 10-16,ROC AUC→1.00].At the cell proliferation level,knockdown of ZNF750 significantly inhibited the proliferation of CESC cell lines(SIHA,C4-II,HCS2,etc.)(CRISPR score <0).Conclusion The upregulation of ZNF750 expression may play a tumor-promoting role in the development of CESC.
