基于TGF-β1/EGR1/NOX4信号通路探讨鳖甲煎丸对人瘢痕疙瘩成纤维细胞的发展研究

【摘要】 目的 探讨鳖甲煎丸通过转化生长因子β1(TGF-β1)/早期生长反应因子1(EGR1)/尼克酰胺腺嘌呤二核苷酸磷酸氧化酶4(NOX4)信号通路调节人瘢痕疙瘩成纤维细胞(KFb)的发展研究。方法 以KFb细胞为研究对象,分别以不同剂量的鳖甲煎丸药物血清培养KFb细胞,分别设置分组为对照组、药物-L组(0.55g/kg鳖甲煎丸)、药物-M组(1.1 g/kg鳖甲煎丸)、药物-H组(2.2 g/kg鳖甲煎丸)以及药物-H+TGF-β1激活剂-SRI-011381组(2.2 g/kg鳖甲煎丸+10 mg/kg SRI-011381)。CCK-8及流式细胞仪检测细胞增殖及凋亡变化;qRT-PCR及Western Blot检测KFb细胞TGF-β1、EGR1、NOX4 mRNA及蛋白表达水平;Transwell检测细胞侵袭。结果 与对照组比较,药物-L组、药物-M组、药物-H组KFb细胞增殖率、侵袭数、TGF-β1、EGR1、NOX4 mRNA及蛋白表达呈浓度依赖性降低,凋亡率呈浓度依赖性增加(P<0.05);与药物-H组比较,药物-H+SRI-011381组KFb细胞KFb细胞增殖率、侵袭数、TGF-β1、EGR1、NOX4 mRNA及蛋白表达增加,凋亡率降低(P<0.05)。结论 鳖甲煎丸可抑制KFb细胞增殖及侵袭,诱导其凋亡,可能与抑制TGF-β1/EGR1/NOX4通路有关。

【Abstract】 Objective To investigate how Biejiajian Wan regulates the development of human keloid fibroblasts(KFb)through the transforming growth factor β1(TGF-β1)/early growth response factor 1(EGR1)/nicotinamide adenine dinucleotide phosphate oxidase 4(NOX4) signaling pathway.Methods Using KFB cells as the research object,KFB cells were cultured with different doses of the drug serum of Baima Jianwu Decoction.The groups were set as control group,drug-L group(0.55g/kg Baima Jianwu Decoction),drug-M group(1.1 g/kg Baima Jianwu Decoction),drug-H group(2.2 g/kg Baima Jianwu Decoction),and drug-H+TGF-β1 activator-SRI-011381 group(2.2 g/kg Baima Jianwu Decoction+10 mg/kg SRI-011381).CCK-8 and flow cytometry were used to detect the changes in cell proliferation and apoptosis;qRT-PCR and Western Blot were used to detect the mRNA and protein expression levels of TGF-β1,EGR1,and NOX4 in KFB cells;Transwell was used to detect cell invasion.Results Compared with control group,the proliferation rate,invasion number,TGF-β1,EGR1,NOX4 mRNA,and protein expression of KFb cells in drug-L group,drug-M group,and drug-H group decreased in a concentration-dependent manner,while the apoptosis rate increased in a concentration-dependent manner(P<0.05);compared with drug-H group,the proliferation rate,invasion number,TGF-β1,EGR1,NOX4 mRNA,and protein expression of KFb cells in drug-H+SRI-011381 group increased,and the apoptosis rate decreased(P<0.05).Conclusion Biejiajian Pills can inhibit the proliferation and invasion of KFb cells and induce their apoptosis.This may be related to the inhibition of the TGF-β1/EGR1/NOX4 pathway.
