富电杂环与半胱氨酸酯的脱硫脱胺偶联反应合成双杂芳基丙酸酯
Synthesis of bisheteroarylpropionates by desulphurisation and deamination coupling reactions of electrically rich heterocycles with cysteine esters
【摘要】 本研究开发了氨基吡唑或吲哚类化合物与半胱氨酸酯盐酸盐通过脱硫、脱氨偶联反应合成双杂芳基丙酸酯的新方法,得出合成双吡唑基丙酸酯和双吲哚基丙酸酯的最优反应条件,制备了多个双杂芳基羧酸酯衍生物,产率中等至优.化合物结构通过核磁共振氢谱1H NMR和核磁共振碳谱13C NMR分析鉴定,并解析了2,2-双(5-氨基-1-苯基-1H-吡唑-4-基)丙酸甲酯的单晶结构,探讨了可能的反应机理.
【Abstract】 A new method was developed for the synthesis of bis(pyrazolyl)propionates by coupling reaction of aminopyrazole or indole analogues with cysteine ester hydrochloride via desulphurisation and deamination. The optimal reaction conditions for synthesizing bis(pyrazolyl) propionate and bis(indolyl) propionate were determined. Several bisheteroarylcarboxylic acid ester derivatives were prepared in moderate to excellent yields. The structures of the compounds were identified by 1H NMR and 13C NMR analyses, and the single crystal structure of methyl 2,2-bis(5-amino-1-phenyl-1H-pyrazol-4-yl)propionate was given. Finally, the possible reaction mechanism was investigated.
