SOX6基因多态性与不同宁都黄鸡品系生长性状的关联性研究

【摘要】 为了研究SOX6基因多态位点对不同宁都黄鸡品系生长性状的影响,试验随机选取3个宁都黄鸡品系(A系、E系、AEB系各80只)共240只健康母鸡为研究对象,利用一代测序和实时荧光定量PCR(RT-qPCR)技术分别筛选SOX6基因的多态位点和检测肌肉组织SOX6基因相对表达量,并利用关联分析和相关分析方法探究多态位点与生长性状(体重、胸肌重、胸肌占比)、SOX6基因相对表达量间的关系。结果表明:SOX6基因上存在3个多态位点,分别为位于13号与14号外显子之间的13_SNP1(rs80610030,A/G,intron variant)、13_SNP2(rs736411303,G/A,intron variant)和位于16号外显子下游3′UTR区的16_SNP1(rs313777635,G/A,3′UTR variant)。A系的13_SNP1(rs80610030,A/G,intron variant)和13_SNP2(rs736411303,G/A,intron variant)位点及3个品系的16_SNP1(rs313777635,G/A,3′UTR variant)位点均符合Hardy-Weinberg平衡(P>0.05)。AEB系、E系的3个多态位点不同基因型的部分或全部生长性状、SOX6基因相对表达量间差异显著(P<0.05)。仅E系中13_SNP1(rs80610030,A/G,intron variant)位点AG基因型的体重、胸肌重和16_SNP1(rs313777635,G/A,3′UTR variant)位点GG基因型的体重与SOX6基因相对表达量呈显著负相关关系(P<0.05)。体重、胸肌重、SOX6基因相对表达量在3个品系间差异显著(P<0.05)。SOX6基因相对表达量分别与体重、胸肌重呈极显著负相关(r=-0.30,P<0.001或r=-0.23,P<0.001),与胸肌占比、宰后15 min肌肉pH值分别呈微弱正相关(r=0.01,P=0.925)和弱正相关(r=0.11,P=0.092)。说明SOX6基因多态位点可作为宁都黄鸡新品系培育的分子标记辅助育种位点。

【Abstract】 In order to study the effects of SOX6 gene polymorphic loci on the growth traits of different Ningdu yellow chicken strains, a total of 240 healthy hens from three Ningdu yellow chicken strains(A strain, E strain, AEB strain, 80 each) were randomly selected as the object of study. The first-generation sequencing and real-time fluorescence quantitative PCR(RT-qPCR) techniques were used to screen the polymorphic loci of SOX6 gene and detect the relative expression of SOX6 gene in muscle tissue. Association analysis and correlation analysis were used to explore the relationship between polymorphic loci, growth traits(body weight, chest muscle weight, chest muscle ratio) and relative expression of SOX6 gene.The results showed that there were three polymorphic loci in SOX6 gene, which were 13_SNP1(rs80610030, A/G, intron variant), 13_SNP2(rs736411303, G/A, intron variant) located between exons 13 and 14, and 16_SNP1(rs313777635, G/A, 3′UTR variant) located in the 3′UTR region downstream of exon 16.The 13_SNP1(rs80610030, A/G, intron variant) and 13_SNP2(rs736411303, G/A, intron variant) loci of the A strain and the 16_SNP1(rs313777635, G/A, 3′UTR variant) loci of the three strains were consistent with Hardy-Weinberg equilibrium(P>0.05). There were significant differences in some or all growth traits and relative expression of SOX6 gene among different genotypes of three polymorphic loci in AEB and E strains(P<0.05). Only the body weight and breast muscle weight of AG genotype at 13_SNP1(rs80610030, A/G, intron variant) locus and the body weight of GG genotype at 16_SNP1(rs313777635, G/A, 3′UTR, variant locus in E strain were significantly negatively correlated with the relative expression of SOX6 gene(P<0.05). There were significant differences in body weight, breast muscle weight and relative expression of SOX6 gene among the three strains(P<0.05). The relative expression of SOX6 gene was significantly negatively correlated with body weight and breast muscle weight(r=-0.30, P<0.001 or r=-0.23, P<0.001), and faintly weakly positively correlated with breast muscle proportion and muscle pH value at 15 min after slaughter(r=0.01, P=0.925) and weakly positively correlated(r=0.11, P=0.092), respectively.It was indicated that the polymonzhic loci of SOX6 gene could serve as molecular marker-assisted breeding loci for the breeding of a new strains of Ningdu yellow chicken.
