SYT13和SYT15在非小细胞肺癌中的表达及对A549细胞增殖迁移能力的影响
The expressions of SYT13 and SYT15 in non-small cell lung cancer and their impacts on the proliferation and migration ability of A549 cells
【摘要】 目的 分析SYT13和SYT15在非小细胞肺癌(NSCLC)中的表达情况,以及它们对A549细胞增殖和迁移能力的影响。方法 选取2018年6月至2021年6月97例NSCLC患者的肿瘤组织和相应的癌旁组织样本,并通过免疫组化方法检测SYT13和SYT15的表达水平。通过Kaplan-Meier生存分析SYT13和SYT15表达对NSCLC患者生存预后的影响,Cox回归分析影响NSCLC患者预后因素。构建了SYT13和SYT15 A549敲低细胞系,通过细胞克隆、细胞划痕和Transwells实验验证了其对A549细胞的增殖、迁移和侵袭能力的影响。结果 NSCLC组织中SYT13的表达在58例(59.79%)(χ~2=22.000,P=0.002),SYT15的表达在58例(59.79%)(χ~2=23.500,P=0.001)显著上调。临床病理特征分析显示,SYT13和SYT15的高表达与患者的肿瘤大小、淋巴结转移、TNM分期和分化程度显著相关(P<0.05)。多因素Cox表明,SYT13(HR=1.674,95%CI:1.036~2.703,P=0.031)和SYT15(HR=1.482,95%CI:1.104~2.137,P=0.009)的高表达与NSCLC患者的较差生存率相关。SYT13高表达组的生存率显著低于低表达组(Logrankχ~2=5.066,P=0.024),而SYT15高表达组的生存率也显著低于低表达组(Logrankχ~2=6.403,P=0.0114)。细胞实验结果表明,敲低SYT13和SYT15显著抑制了A549细胞的增殖、迁移和侵袭能力(P<0.01)。结论 SYT13和SYT15在非细小肺癌患者体内高表达,且与临床病理特征和预后关联,可作为联合检测标志基因。
【Abstract】 Objective To investigate the expressions of synaptotagmin-13(SYT13) and SYT15 in non-small cell lung cancer(NSCLC) and their impacts on the proliferation and migration abilities of A549 cells.Methods Tumor and adjacent non-tumor tissue samples were collected from 97 NSCLC patients admitted in the Guangzhou Twelfth People’s Hospital from June 2018 to June 2021.The expression levels of SYT13 and SYT15 were detected via immunohistochemistry.Kaplan-Meier survival analysis was conducted to assess the effect of SYT13 and SYT15 on the survival of NSCLC patients.Cox regression analysis was performed to identify prognostic factors affecting NSCLC patients.SYT13 and SYT15 knockdown cell lines were constructed in A549 cells.Cell proliferation, migration, and invasion abilities were evaluated through colony formation, wound healing, and Transwell assays, respectively.Results SYT13 expression was significantly upregulated in 58 cases(59.79%)(χ~2=22.000,P=0.002), and SYT15 expression was significantly upregulated in 58 cases(59.79%)(χ~2=23.500, P=0.001) of NSCLC tissues. Clinicopathological analysis revealed that high expression of SYT13 and SYT15 was significantly correlated with tumor size, lymph node metastasis, TNM stage, and degree of differentiation(P<0.05). Multivariate Cox regression analysis indicated that high expression of SYT13(HR=1.674, 95%CI: 1.036-2.703,P=0.035)and SYT15(HR=1.482, 95%CI: 1.104-2.137,P=0.009) was associated with poor survival in NSCLC patients. Survival rates in the high SYT13 expression group were significantly lower than those in the low expression group(Log-rank χ~2=5.066, P=0.024), and similarly, survival rates in the high SYT15 expression group were significantly lower than those in the low expression group(Log-rank χ~2=6.403, P=0.0114). Cellular experiments demonstrated that knockdown of SYT13 and SYT15 significantly inhibited A549 cell proliferation, migration, and invasion abilities(P<0.01).Conclusion SYT13 and SYT15 are highly expressed in NSCLC patients and are associated with clinicopathological characteristics and prognosis.They can serve as combined diagnostic markers.
