具鳞水柏枝的HPLC指纹图谱分析及有效成分含量测定
HPLC Fingerprint Analysis and Active Ingredient Content Determination of Myricaria squamosa Desv.
【摘要】 采用Spursil C18色谱柱(250 mm×4.6 mm, 5μm),以乙腈-0.3%磷酸水溶液为流动相进行梯度洗脱,流速1.0 mL·min-1,洗脱时间110 min,检测波长210、254 nm,柱温30℃,进样体积10μL,建立了具鳞水柏枝HPLC指纹图谱,并对有效成分含量进行了测定。结果表明,49批次具鳞水柏枝的HPLC指纹图谱有18个共有峰;经与标准品比对,指认出其中8个共有峰,分别为没食子酸、没食子酸甲酯、香草酸、咖啡酸、阿魏酸、芦丁、槲皮素-3-葡萄糖醛酸苷、槲皮苷,其含量分别为0.001 0~0.017 0、0.002 0~0.017 5、0.002 8~0.042 0、0.001 8~0.023 5、0.001 3~0.022 0、0.061 3~0.377 3、0.019 3~0.143 5、0.004 3~0.058 5 mg·g-1;49批次具鳞水柏枝的HPLC指纹图谱与对照指纹图谱相似度为0.893~0.988,化学成分基本相似;49批次具鳞水柏枝分为2类,质量差异标志物为10号峰(阿魏酸)、9号峰、14号峰(槲皮素-3-葡萄糖醛酸苷)、16号峰、5号峰(香草酸)、8号峰、12号峰(芦丁)。该方法简便、专属性强、稳定、可重复,可为具鳞水柏枝药材的鉴别与质量评价提供参考。
【Abstract】 Using a Spursil C18 chromatographic column(250 mm×4.6 mm, 5 μm),acetonitrile-0.3% phosphoric acid aqueous solution as a mobile phase for gradient elution with following parameters: the flow rate of 1.0 mL·min-1,the elution time of 110 min, the detection wavelengths of 210 and 254 nm, the column temperature of 30 ℃,and the injection volume of 10 μL,we established an HPLC fingerprint of Myricaria squamosa Desv.,and determined the contents of active ingredients.The results show that there are 18 common peaks in the HPLC fingerprint of 49 batches of Myricaria squamosa Desv..Compared with standard sample, 8 common peaks are identified, namely gallic acid, methyl gallate, vanillic acid, caffeic acid, ferulic acid, rutin, quercetin-3-glucuronide, and quercetin, with contents of 0.001 0-0.017 0,0.002 0-0.017 5,0.002 8-0.042 0,0.001 8-0.023 5,0.001 3-0.022 0,0.061 3-0.377 3,0.019 3-0.143 5,0.004 3-0.058 5 mg·g-1,respectively.The similarity between the HPLC fingerprint of 49 batches of Myricaria squamosa Desv.and the control fingerprint is 0.893-0.988,and the chemical ingredients are similar.The 49 batches of Myricaria squamosa Desv.are divided into two categories, with quality difference markers being peak 10(ferulic acid),peak 9,peak 14(quercetin-3-glucuronide),peak 16,peak 5(vanillic acid),peak 8,and peak 12(rutin).This method is simple, highly specific, stable, and reproducible, which provides a reference for the identification and quality assessment of Myricaria squamosa Desv.medicinal materials.
