抑郁症和肥胖:一项两样本孟德尔随机化分析

【摘要】 目的 采用孟德尔随机化法,以单核苷酸多态性(P<5×10-8)为工具变量,探究抑郁症对体重指数的因果作用。方法 从欧洲生物信息研究所获取抑郁症、体重指数的遗传关联数据。采用MR-Egger回归、加权中位数法、随机效应逆方差加权法、简单模型和加权模型,使用R语言中的TwoSampleMR包进行孟德尔随机化分析和结果可视化。结果 在不同的分析中,基因预测的抑郁症与体重指数呈正相关。在逆方差加权分析中,抑郁症导致体重指数的OR为1.44(95%CI,1.16~1.71,P=0.007),且不存在异质性和水平多效性。结论 本研究提供了抑郁症与体重指数风险增加之间的因果关系的证据,但其具体的生物学机制还需要进一步的研究。

【Abstract】 Objective To explore the causal effect of depression on body mass index by using Mendelian Randomization. Method and single nucleotide polymorphism(P<5×10-8) as instrumental variable. Methods:Genetic association data for depression and body mass index were obtained from the European Bioinformatics Institute. Mendelian randomization analysis was carried out by MR-Egger regression,weighted median method,random effect inverse variance weighted method,simple model and weighted model. Using the TwoSampleMR package in R software for Mendelian randomization analysis and result visualization. Results In separate analyses,genetically predicted depression was positively associated with body mass index. In inverse variance weighted analysis,OR for depression leading to body mass index were 1.44(95% CI,1.16-1.71,P=0.007),with no heterogeneity and horizontal pleiotropy. Conclusions This study provides evidence of a causal relationship between depression and increased risk of body mass index,but the specific biological mechanisms require further investigation.
