帕金森病蛋白2在瘢痕疙瘩中的表达及意义

【摘要】 目的 研究人正常皮肤和瘢痕疙瘩组织中帕金森病蛋白2(PARK2)的表达情况及意义。方法 分别对2种组织进行转录组测序,定量分析PARK2 mRNA表达情况,分析PARK2相关的基因,以及富集的相关通路情况,以及相互作用蛋白网络,最后对2种组织进行免疫组织化学并对图像定量分析。结果瘢痕疙瘩组织中PARK2在RNA水平表达情况,与正常皮肤组织相比统计发现有显著差异(P<0.05)。差异表达上调基因、自噬相关基因和PARK2负相关基因交集获得77个基因。其中排名第一核心基因的是PLOD2,可能是PARK2的相关联作用蛋白。定量分析2种组织免疫组化切片图像发现瘢痕疙瘩中的PARK2蛋白表达存在低表达。结论 PARK2可能是瘢痕疙瘩的一个新治疗靶点。

【Abstract】 Objective This study investigates the expression and significance of PARK2 in normal human skin and keloid tissue. Methods Transcriptome sequencing was performed on the two tissues to quantitatively analyze PARK2 mRNA expression, PARK2-related genes, enriched pathways, and interacting protein networks. Finally, immunohistochemistry was performed on the two tissues and the images were quantitatively analyzed. Results The expression of PARK2 at the RNA level in keloid tissue was significantly different from that in normal skin tissue(P<0.05). A total of 77 genes were identified through the intersection of differentially upregulated genes, autophagyrelated genes, and PARK2-negative genes. PLOD2 was the leading gene, potentially a PARK2-related protein. Quantitative analysis of immunohistochemical sections from both tissues revealed low PARK2 protein expression in keloids. Conclusion PARK2 may be a novel therapeutic target for keloids.
