罗布麻叶不同溶剂提取部分指纹图谱研究
Fingerprint Analysis of Extracts from Apocynum venetum L. Using Different Solvents
【摘要】 目的:将罗布麻叶药材用不同极性溶剂处理,建立各部分的指纹图谱,比较罗布麻叶不同提取部分中的化学成分的差异,为罗布麻叶的质控提供科学依据。方法:建立12批罗布麻叶的不同溶剂提取部分的高效液相色谱指纹图谱。结合相似度评价、聚类分析、主成分分析进行不同批次罗布麻叶质量评价。结果:12批罗布麻叶80%甲醇超声、60%乙醇回流、水提醇沉上清三个提取部位分别确定26、25、21个共有峰,共指认绿原酸、金丝桃苷等8种成分;通过CA和PCA分析可将罗布麻叶药材分为2类。结论:本实验建立的指纹图谱可以较为全面地反映罗布麻叶中化学成分的分布,可为罗布麻叶药材质量控制提供参考依据。
【Abstract】 Objective: To process Apocynum venetum leaves with solvents of different polarities, establish fingerprint profiles for each fraction, compare the differences in chemical composition among the different extract fractions of Apocynum venetum leaves, and provide a scientific basis for quality control of Apocynum venetum leaves. Methods: High-performance liquid chromatography(HPLC) fingerprint profiles were established for the different solvent extract fractions of 12 batches of Apocynum venetum leaves. Quality evaluation of the different batches was performed by combining similarity assessment, cluster analysis(CA), and principal component analysis(PCA). Results: For the 12 batches of Apocynum venetum leaves, 26, 25, and 21 common peaks were identified in the 80% methanol ultrasonic extract, 60% ethanol reflux extract, and water extract-alcohol supernatant fractions, respectively. Eight components, including chlorogenic acid and hyperoside, were identified. CA and PCA analysis classified the Apocynum venetum leaf samples into two categories. Conclusion: The fingerprint profiles established in this experiment can comprehensively reflect the distribution of chemical components in Apocynum venetum leaves and may serve as a reference for the quality control of Apocynum venetum leaf material.
