绿色吲哚C3-氨甲基化综合实验教学设计

【摘要】 针对当前本科有机化学实验教学中绿色化学理念融入不足、学生综合实验技能训练薄弱及创新能力培养欠缺等问题,设计并开发了一项绿色无催化吲哚C3-氨甲基化综合教学实验。该实验以可循环利用的1,2-二氯乙烷为溶剂,整合了半微量合成、薄层色谱(TLC)监测、柱层析分离等核心操作环节,系统训练学生的综合实验能力。教学实践(共5组10名学生)结果显示:目标产物平均收率达82±5%,90%学生规范掌握了无水无氧操作技术,100%学生能独立完成TLC监测。该实验将绿色化学原则贯穿于“合成-监测-分离-表征”全流程,并提供适用于基础实验室条件的替代表征方案,有效提升了学生的绿色化学意识和综合实验技能。

【Abstract】 To address the prevailing challenges in undergraduate organic chemistry experiments—insufficient integration of green chemistry principles, inadequate training in comprehensive experimental skills, and lack of emphasis on fostering innovative capabilities—this study designed and implemented a comprehensive teaching experiment featuring green, catalyst-free C3-aminomethylation of indole. Utilizing recyclable 1,2-dichloroethane as a solvent, the experiment integrates core techniques including semi-microscale synthesis, thin-layer chromatography(TLC) monitoring, and column chromatography separation, thereby systematically enhancing students’ comprehensive experimental proficiency. Implementation with 5 groups(10 students total) demonstrated an average product yield of 82±5%, with 90% of students mastering standardized air-free techniques and 100% achieving proficiency in TLC monitoring. Crucially, green chemistry principles are embedded throughout the “synthesis-monitoring-separation-characterization” workflow, and offer accessible characterization methods tailored to basic laboratory conditions. This approach has effectively strengthened students’ awareness of green chemistry and improved their comprehensive experimental skills.
