药用植物甜叶菊茎段离体再生体系的建立
Establishment of Plant Regeneration System Induced by Stem Bud Seedling of Medicinal Plant Stevia rebaudiana
【摘要】 以甜叶菊带腋芽的茎段为试材,研究了不同消毒条件、不同浓度组合的植物生长调节剂以及不同养护方式对甜叶菊组培快繁的影响,以期建立完整的植株再生体系。结果表明,茎段消毒的最佳组合为75%乙醇处理10 s,0.1%氯化汞处理4 min,此条件下污染率为10.7%,存活率为89.3%;最佳初代培养基为MS+1.5 mg/L 6-BA+0.2 mg/L NAA,一次诱导率为93.3%;最佳增殖培养基为MS+0.5 mg/L 6-BA+0.05 mg/L NAA,30 d增殖系数达7.50;MS+0.5 mg/L 6-BA+0.5 mg/L NAA是理想的壮苗培养基,此条件下组培苗生长状态较好,利于生根;壮苗培养后的试管苗在MS+0.5 mg/L 6-BA+0.5 mg/L NAA的培养基中生根效果最好,28 d后,生根率达96.5%,单株苗平均生根数达11条;生根苗经25 mg/L生根粉处理后,移入3∶1∶1的园土、红土和细沙基质中栽培,成活率达96.0%。本研究建立的甜叶菊离体再生体系,为工厂化育苗和种质资源保育提供思路和帮助。
【Abstract】 Taking stems with axillary buds of Stevia rebaudiana as test materials, the effects of different disinfection conditions, different concentrations of plant growth regulators and different maintenance methods on tissue culture and rapid propagation of Stevia rebaudiana were studied in order to establish a complete plant regeneration system. The results showed that the best combination of stem disinfection was 75% alcohol treatment for 10 s and 0.1% mercuric chloride treatment for 4 min. Under this condition, the pollution rate was 10.7% and the survival rate was 89.3%. The optimal primary medium was MS +1.5 mg/L 6-BA +0.2 mg/L NAA, and the primary induction rate was 93.3%. The best proliferation medium was MS+0.5 mg/L 6-BA+0.05 mg/L NAA, and the proliferation coefficient was 7.50 after 30 days. MS+0.5 mg/L 6-BA+0.5 mg/L NAA is the ideal medium for strong seedlings, under this condition, the growth state of tissue culture seedlings is better, which is beneficial to rooting; the rooting effect of the test-tube seedlings cultured with strong seedlings was the best in the medium of MS+0.5 mg/L 6-BA+0.5 mg/L NAA. After 28 days, the rooting rate reached 96.5%, and the average number of roots per plant reached 11; after the seedlings were treated with 25 mg/L rooting powder, they were transferred into3:1:1 garden soil, red soil and fine sand matrix for cultivation, and the survival rate reached 96.0%. The regeneration system of the stem segments of Stevia rebaudiana in vitro that has been optimized in the current study will provide ideas and help for industrial seedling and germplasm resources conservation.
