鲁米诺/二氧化硫脲流动注射化学发光灵敏检测亚硝酸钠

【摘要】 亚硝酸钠(NaNO2)作为食品着色剂和防腐剂被广泛使用,但是过量摄入易诱发胃癌等疾病,急性过量摄入还可引发高铁血红蛋白症,导致严重急性中毒症状。因此,快速准确检测NaNO2浓度对于避免急性中毒及监控长期过量摄入所致疾病具有重要意义。鉴于化学发光方法具有设备小型化、操作简便和灵敏度高等优点,结合二氧化硫脲作为鲁米诺共反应剂可提高化学发光体系的稳定性和选择性,本研究建立了鲁米诺/二氧化硫脲流动注射化学发光定量分析NaNO2的方法。在体系中加入不同浓度的NaNO2后,NaNO2与溶液中的二氧化硫脲发生反应,消耗鲁米诺的共反应物,导致鲁米诺/二氧化硫脲的化学发光信号不同程度降低。优化了溶液pH值和蠕动泵流速的实验条件。在最佳实验条件下,本方法检测NaNO2的线性范围为10 nmol/L~5 mmol/L,相关系数(r)为0.9971,检出限(S/N=3)为3.5 nmol/L。对10μmol/L NaNO2连续9次进样检测,发光信号的相对标准偏差为4.36%,常见共存物质对NaNO2的检测无干扰,表明本方法具有良好的重现性和较强的抗干扰能力。

【Abstract】 Sodium nitrite(NaNO2) is widely used as a food additive. Long-term excessive intake of NaNO2 may induce diseases such as gastric cancer, while acute overconsumption can lead to methemoglobinemia which shows severe acute poisoning symptoms. Therefore, rapid and accurate detection of NaNO2 is crucial for preventing acute poisoning as well as monitoring long-term excessive intake-induced diseases. Chemiluminescence(CL) is a method which has advantages of compact instrumentation, simplicity of detection, and high sensitivity. Combining with the benefits of using thiourea dioxide as a luminol co-reagent to enhance the stability and selectivity of the CL system, a luminol/thiourea dioxide flow injection CL system was employed for quantitative analysis of NaNO2. The significant reduction in luminescence upon addition of NaNO2 demonstrated the feasibility of this analytical method. Optimal detection conditions were further established by optimizing solution p H value and flow rate of peristaltic pump. The system was proved suitable for quantifying NaNO2 from 10 nmol/L to 5 mmol/L, with a detection limit(S/N=3) of3.5 nmol/L. The linear correlation coefficient(r) was 0.9971. A relative standard deviation of 4.36% was obtained for nine consecutive injections of 10μmol/L NaNO2, indicating a good reproducibility. Furthermore, the addition of six representative interferents showed no significant effect on CL intensity, indicating a good selectivity.
