互花米草提取物对白羽肉鸡免疫功能的影响
Effect of Spartina alterniflora Extract on Immune Function of White-Feather Broilers
【摘要】 试验旨在研究互花米草提取物对白羽肉鸡免疫功能的影响。将288只1日龄白羽肉公鸡随机分成6组,每组6个重复,每个重复8只鸡,试验期42 d,试验分为1~21日龄和22~42日龄2个阶段。空白对照组(CK组)饲喂基础饲粮,试验组分别在基础饲粮中添加500(S1组)、1 000(S2组)、1 500(S3组)、2 000(S4组) mg/kg互花米草提取物,阳性对照组(PC组)在基础饲粮中添加200 mg/kg金霉素。测定血清免疫指标、免疫器官指数、肠道白细胞介素和紧密连接蛋白相对表达量,进行单因素方差分析。结果表明:21日龄时,S2组血清免疫球蛋白A(IgA)含量显著高于CK、S4、PC组,免疫球蛋白G(IgG)含量显著高于S4组和PC组,CK组干扰素-γ(IFN-γ)含量显著高于除S1组以外的其他4组。42日龄时,S3组血清免疫球蛋白M(IgM)含量显著高于S1组和PC组,CK组和S1组IFN-γ含量显著高于其他4组。互花米草提取物添加量为2 000 mg/kg时,胸腺指数最高,肠道白细胞介素10(IL-10)mRNA相对表达水平显著高于对照组;添加量为500 mg/kg时,肠道ZO-1、Occludin蛋白表达显著高于对照组。综上所述,互花米草提取物可以增强白羽肉鸡免疫功能。
【Abstract】 This experiment was conducted to study the effects of Spartina alterniflora extract on immune function of white feather broilers. A total of 288 1-day-old white-feathered male broilers were randomly divided into six groups with six replicates per group and eight chickens per replicate. The experiment lasted for 42 d. The experiment was divided into two stages: 1-21 d old and 22-42 d old. Broilers in the blank control group(CK group) were fed a basal diet. Broilers in the experimental groups were fed the basal diet supplemented with 500(S1 group), 1 000(S2 group), 1 500(S3 group) and 2 000 mg/kg(S4 group) Spartina alterniflora extract, respectively. Broilers in the positive control group(PC group) were fed the basal diet supplemented with 200 mg/kg chlortetracycline. Serum immune index, immune organ index, relative expression of intestinal interleukin and tight junction protein were measured, and single factor analysis of variance was performed. The results showed that at 21 d of age, the serum IgA contents of S2 group were significantly higher than those of CK, S4 and PC groups, the IgG content was significantly higher than that of S4 and PC groups, and the IFN-γ content of CK group was significantly higher than that of the other four groups except S1. At 42 d of age, serum IgM content in S3 group was significantly higher than that in S1 and PC groups, and IFN-γ content in CK and S1 groups was significantly higher than that in the other four groups. When the addition amount of Spartina alterniflora extract was 2 000 mg/kg, the thymus index was the highest, and the expression level of intestinal IL-10 mRNA was significantly higher than that of the control group. When the addition amount was 500 mg/kg, the expression of intestinal ZO-1 and Occludin protein was significantly higher than that of the control group. Collectively, Spartina alterniflora extract can enhance the immune function of white feather broilers.
