基于转录组学解析丁氏乌蔹莓软膏治疗肛瘘术后创面的作用机制
Analysis of the action mechanism of DINGs’ Wulianmei Ointment on postoperative wound of anal fistula based on transcriptomics
【摘要】 目的:利用基因测序技术,探究丁氏乌蔹莓软膏治疗肛瘘术后创面的潜在作用机制。方法:10只C57BL/6J小鼠随机分为模型组和乌蔹莓软膏组,用葡聚糖硫酸钠联合不锈钢铁丝构建肛瘘术后创面模型,造模成功后,分别予0.9%氯化钠溶液、0.9%氯化钠溶液+乌蔹莓软膏换药。11 d后取样,HE染色观察肉芽组织病理学变化,转录组学分析相关作用靶点。结果:与模型组比较,乌蔹莓软膏组肌肉纤维明显,炎性细胞浸润明显减少,结构层次分明,可见新生的毛细血管。转录组学结果显示,PI3K-Akt信号通路、NOD受体/TNF/IL-17信号通路是乌蔹莓软膏治疗术后创面的潜在作用机制。结论:丁氏乌蔹莓软膏可通过激活促修复轴、抑制炎症相关通路协同治疗肛瘘术后创面。
【Abstract】 Objective: To investigate the potential action mechanism of DING’s Wulianmei Ointment(CJC) in the treatment of postoperative wound of anal fistula by gene sequencing technology. Methods: Ten C57 BL/6J mice were randomly divided into model group and CJC group. The postoperative wound was induced by dextran sulfate sodium combined with stainless steel wire. After the model was successfully established, the two groups were treated with normal saline, normal saline+CJC, respectively. After 11 days, samples were taken for HE staining to observe the pathological changes of granulation tissue and transcriptome analysis of related targets. Results: Compared with the model group, the muscle fibers and inflammatory cells were obviously reduced in the CJC group, the structure of the CJC group was well defined, and new capillaries were observed. The results of transcriptome analysis showed that PI3K-Akt signaling pathway and NOD receptor/TNF/IL-17 signaling pathway were the potential mechanisms of CJC in the treatment of postoperative wound. Conclusion: CJC can treat the postoperative wound of anal fistula by activating the pro-repair axis and inhibiting the inflammation-related pathways.
