基于量子点技术的牛结节性皮肤病病毒P32抗体免疫层析检测试纸的制备

【摘要】 为了建立一种快速、高灵敏度的牛结节性皮肤病病毒(lumpy skin disease virus, LSDV)P32蛋白抗体的免疫层析检测方法,本研究基于量子点(quantum dots, QDs)荧光微球标记技术制作免疫层析试纸条,对QDs与P32蛋白的偶联条件、免疫层析试纸条的最佳制备参数进行了优化,并对试纸条的特异性、灵敏性、重复性与稳定性进行了检测,最后与商品化检测试剂盒的检测结果进行了比对。结果表明,该方法与牛病毒性腹泻病毒(bovine viral diarrhea virus, BVDV)、小反刍兽疫病毒(peste des petits ruminants virus, PPRV)、牛冠状病毒(bovine coronavirus, BCoV)、禽支原体(mycoplasma gallisepticum, MG)等常见病原无交叉反应,特异性良好;最低可检测1∶160倍稀释的LSDV阳性血清;保存120 d的试纸条仍具有良好的检测效果,稳定性优。与商品化检测试剂盒检测方法对比,本方法符合率达到86.00%,且敏感性更高,适用于临床样本的早期筛查。

【Abstract】 In order to establish a rapid and highly sensitive immunochromatographic method for the detection of lumpy skin disease virus(LSDV) P32 protein antibodies, this study had developed immunochromatographic strips based on quantum dot(QDs) fluorescent microsphere labeling technology.The conjugation conditions of QDs with P32 protein and the optimal preparation parameters of the immunochromatographic strips were optimized.The specificity, sensitivity, reproducibility, and stability of the strips were evaluated, and the results were compared with those of a commercial detection kit.The results showed that this me-thod exhibited no cross-reaction with common viruses such as bovine viral diarrhea virus(BVDV),peste des petits ruminants virus(PPRV),bovine coronavirus(BCoV),and mycoplasma gallisepticum(MG),indicating good specificity.It could detect LSDV-positive serum diluted up to 1∶160,demonstrating high sensitivity.The strips maintained good detection performance after 120 days of storage, confirming excellent stability.Compared with the commercial detection kit, this method achieved a concordance rate of 86.00% and showed higher sensitivity, making it suitable for early screening of clinical samples.
