基于Tfh细胞功能调控探讨穿山龙薯蓣皂苷元改善自身免疫性甲状腺炎的机制

【摘要】 目的 探究穿山龙薯蓣皂苷元对自身免疫性甲状腺炎(Autoimmune thyroiditis,AIT)小鼠模型中甲状腺过氧化物酶抗体(TPOAb)、甲状腺球蛋白抗体(TgAb)、游离三碘甲状腺原氨酸(FT3)、游离甲状腺素(FT4)、促甲状腺激素(TSH)的调节作用,以及滤泡辅助性T细胞(Tfh)相关因子B淋巴细胞瘤转录因子6(BCL-6)、白细胞介素(IL)-21的调控机制。方法 采用浓度为0.064%的碘化钠溶液(高碘水)联合甲状腺球蛋白与佐剂免疫法构建AIT模型,模型验证成功后随机分为模型对照组M、阳性对照组P及薯蓣皂苷元低剂量组L、薯蓣皂苷元高剂量组H。每日灌胃相应药物或对照溶液,连续4周。通过ELISA检测血清AIT相关抗体、激素及Tfh相关因子水平,荧光定量PCR检测Tfh相关因子mRNA表达。结果 薯蓣皂苷元显著下调AIT模型小鼠血清TgAb、TPOAb、TSH水平,上调FT3、FT4水平,同时降低IL-21、BCL-6的mRNA表达,且呈剂量依赖性,干预效果与剂量正相关。结论 穿山龙薯蓣皂苷元通过调控Tfh细胞相关因子IL-21、BCL-6调节AIT相关抗体及激素表达,为AIT的免疫干预提供潜在策略。

【Abstract】 Objective To investigate the regulatory effects of diosgenin from Dioscorea nipponica on autoimmune thyroiditis(AIT)-related antibodies [thyroperoxidase antibody(TPOAb), thyroglobulin antibody(TgAb)], hormones [free triiodothyronine(FT3), free thyroxine(FT4), thyroid-stimulating hormone(TSH)], and the regulatory mechanism of T follicular helper(Tfh) cell-associated factors [Bcell lymphoma 6(BCL-)6, interleukin-21(IL-21)] in an AIT mouse model. Methods The AIT model was established by using highiodine water combined with thyroglobulin and adjuvant immunization. After successful model verification, the rats were randomly divided into the model control group, positive control group, and diosgenin low-dose group and diosgenin high-dose group. Corresponding drugs or control solutions were administered by gavage daily for 4 consecutive weeks. The levels of AIT-related antibodies, hormones and Tfh-related factors in serum were detected by ELISA, and the mRNA expression of Tfh-related factors was detected by fluorescent quantitative PCR. Results Diosgenin significantly downregulated the serum levels of TgAb, TPOAb, and TSH, and upregulated the levels of FT3 and FT4 in AIT model mice. Meanwhile, it reduced the mRNA expressions of IL-21 and BCL-6 in a dosedependent manner, with the intervention effect positively correlated with the dose. Conclusion Diosgenin from Dioscorea nipponica ameliorates AIT by modulating Tfh cell-associated factors IL-21 and BCL-6, thereby regulating antibody and hormone secretion. This study provides a potential therapeutic strategy for AIT immune intervention.
