PD相关蛋白DJ-1及其突变体L166P在鱼藤酮诱导的细胞损伤的作用研究
Study on the role of PD related protein DJ-1 and its mutant L166P in rotenone induced cell damage
【摘要】 目的 通过分析鱼藤酮处理预先转染编码DJ-1、DJ-1突变蛋白L166P及DJ-1敲除(KD)质粒的SH-SY5Y细胞的损伤情况,探讨DJ-1在鱼藤酮诱导SH-SY5Y细胞损伤中的保护作用。方法 构建了编码DJ-1、L166P及DJ-1 KD的质粒,并将其转染至SH-SY5Y细胞。随后,所有转染后的SH-SY5Y细胞均接受鱼藤酮处理,并对不同转染组的细胞损伤情况进行分析。结果 DJ-1 KD转染组和L166P转染组在鱼藤酮处理后,SH-SY5Y细胞的损伤程度明显高于DJ-1转染组。结论 DJ-1突变蛋白L166P由于突变影响其二聚体的形成,进而影响其细胞定位,导致其无法有效发挥对鱼藤酮诱导的氧化损伤的保护作用。
【Abstract】 Objective This study aims to investigate the impact of DJ-1, specifically the L166P mutant, and DJ-1 knockdown(KD) on the cellular damage inflicted on SH-SY5Y cells following exposure to rotenone, thereby elucidating the role of DJ-1 in mediating rotenone-induced cell damage. Methods Plasmids encoding DJ-1, the L166P mutant, and DJ-1 KD were constructed and subsequently transfected into SH-SY5Y cells. Following transfection, the cells were uniformly exposed to rotenone, and the extent of cellular damage across the different transfection groups was assessed. Result indicated that SH-SY5Y cells in the DJ-1 KD and L166P transfection groups exhibited significantly greater damage after rotenone treatment compared to those in the DJ-1 transfection group. Conclusion The L166P mutant of DJ-1 does not confer protection against oxidative damage induced by rotenone, likely due to the mutation disrupting dimer formation and altering subcellular localization.
