LncRNA CD36-005对大鼠子宫内膜蜕膜化的作用机制研究

【摘要】 目的 探究lncRNA CD36-005与大鼠子宫内膜蜕膜化过程的相关性及其作用机制。方法 复制早期妊娠大鼠子宫、人工诱导蜕膜化子宫模型,通过实时荧光定量聚合酶链反应(qRT-PCR)检测lncRNA CD36-005表达水平。以体外诱导蜕膜化的内膜基质细胞为体外模型,分别过表达及干扰lncRNA CD36-005的表达后,再通过qRT-PCR检测蜕膜化标志基因的表达水平。结果 妊娠第5~8天的雌性大鼠子宫中lncRNA CD36-005相对表达量较妊娠第1~4天低(P<0.05)。在妊娠第6~8天的子宫中,子宫着床位点的lncRNA CD36-005相对表达量低于非着床位点(P<0.05)。人工诱导蜕膜化子宫组织及蜕膜化的内膜基质细胞中lncRNA CD36-005相对表达量均显著下降(P<0.05)。在蜕膜化的内膜基质细胞中过表达lncRNA CD36-005可显著抑制Hand2、Prl8a2相对表达量(P<0.05),反之,lncRNA CD36-005干扰后,Hand2和Prl8a2相对表达量显著升高(P<0.05)。结论 LncRNA CD36-005在大鼠早期妊娠子宫、人工诱导蜕膜化子宫和蜕膜化的内膜基质细胞中低表达,并对子宫内膜蜕膜化有抑制作用。

【Abstract】 Objective To explore the association between lncRNA CD36-005 and endometrial decidualization, as well as the role of lncRNA CD36-005 in decidualization of the rat endometrium. Methods Models of early pregnancy and artificially induced decidualization in rat uteri were established. The expression level of lncRNA CD36-005 was detected by quantitative real-time polymerase chain reaction(qRT-PCR). An in vitro model of decidualization was established using endometrial stromal cells. Following overexpression or knockdown of lncRNA CD36-005, the expression levels of decidualization marker genes were assessed by qRT-PCR. Results The relative expression level of lncRNA CD36-005 in the uterine tissues of female rats on gestation days 5-8 was significantly lower than that on days 1-4(P < 0.05). On gestation days 6-8, the expression of lncRNA CD36-005 at implantation sites was significantly lower than that at non-implantation sites(P < 0.05). In both artificially induced decidualized uterine tissues and decidualized endometrial stromal cells, lncRNA CD36-005 expression was markedly reduced(P < 0.05). Overexpression of lncRNA CD36-005 in decidualized stromal cells significantly inhibited the expression of Hand2 and Prl8a2(P < 0.05), whereas knockdown of lncRNA CD36-005 significantly increased the expression of these two genes(P < 0.05). Conclusion LncRNA CD36-005 is lowly expressed in the uterus during early pregnancy, in artificially induced decidualized uteri, and in decidualized endometrial stromal cells of rats, and it exerts an inhibitory effect on endometrial decidualization.
