RFRP-3对小鼠卵泡发育和类固醇激素合成的影响
Effects of RFRP-3 on follicular development and steroid synthesis in mice
【摘要】 旨在探究RFRP-3对小鼠卵泡发育和类固醇激素合成的影响。本试验将40只6周龄KM雌鼠分为4组,分别为0(对照组)、100、500、2 000 ng/d组,每日腹腔注射对应剂量RFRP-3,试验期共7 d。通过测量卵巢湿质量,计算并统计各组卵巢系数;通过HE染色,分析不同浓度RFRP-3对小鼠卵泡发育的影响;通过Western blot检测小鼠卵巢内细胞凋亡、类固醇合成酶和卵母细胞分泌因子的蛋白表达;通过放射免疫法检测小鼠血清中孕酮(P4)和雌二醇(E2)的浓度。结果表明,与对照组相比,各试验组小鼠体质量没有显著差异;500和2 000 ng/d处理组卵巢系数与对照组相比极显著下降(P<0.01);与对照组相比,500和2 000 ng/d处理组次级卵泡和窦卵泡数极显著下降(P<0.01),闭锁卵泡数极显著升高(P<0.01);500和2 000 ng/d处理组Bax和Caspase3蛋白表达与对照组相比显著升高(P<0.05),Bcl-2蛋白表达对照组相比极显著下降(P<0.01);与对照组相比,500和2 000 ng/d处理组3βHSD、StAR、CYP11a1、CYP17a1蛋白表达极显著降低(P<0.01);与对照组相比,500 ng/d处理组E2浓度显著下降(P<0.05),2 000 ng/d处理组E2浓度极显著下降(P<0.01),各试验组P4浓度没有显著变化;500和2 000 ng/d处理组BMP15和GDF9的基因与蛋白表达与对照组相比均显著下降(P<0.05)。综上,RFRP-3可通过调控小鼠卵巢内凋亡因子、卵母细胞分泌因子和类固醇合成酶的表达,从而抑制小鼠卵泡发育和类固醇激素的分泌。本试验为RFRP-3对哺乳动物卵泡发育和类固醇激素合成的调控机理提供了理论依据。
【Abstract】 Elucidate the influence of RFRP-3 on follicular development and steroid hormone synthesis in mice.Forty 6-week-old female KM mice were randomly allocated into four groups,namely the 0(control),100,500 and 2 000 ng/d treatment groups.Each group received daily intraperitoneal injections of RFRP-3 for a duration of 7 days.The ovarian coefficients were calculated and recorded by measuring the wet weight of the ovaries.The effects of RFRP-3 at different concentrations on follicular maturation in mice were investigated through HE staining.Western blot analysis was employed to assess the expression of apoptosis-relaled proteins,steroidogenic enzymes,and oocyte secretion factors within the mouse ovaries.The concentrations of progesterone(P4) and estradiol(E2) in the serum of the mice were determined using radioimmunoassay techniques.The results revealed no significant differences in body weight between the experimental groups and the control.However,the ovarian coefficients in the 500 and 2 000 ng/d treatment groups were significantly reduced when compared to the control(P<0.01).A marked decrease in the numbers of secondary and antral follicles was observed in the 500 ng/d and 2 000 ng/d groups(P<0.01),concomitant with a substantial increase in atretic follicles(P<0.01).A significant decline in follicle count was also noted in these groups(P<0.01).Compared to the control,the protein expression of Bax and Caspase3 was Substantially upregulated in the 500 and 2 000 ng/d treatment groups(P<0.05),while the expression of Bcl-2 protein was significantly downregulated(P <0.01).Similarly;the protein expression of 3PHSD,SCAR,CYP11a1,and CYP17a1 was significantly decreased at the 500and 2 000 ng/d groups(F <0.01).E2 concentrations were moderately reduced in the 500 ng/d group(P<0.05) and significantly decreased in the 2000 ng/d group(P<0.01),whereas P4 levels remained unchanged across all experimental groups.Notably,the expression of BMP 15 and GDF9at both the gene and protein levels was markedly attenuated in the 500 and 2 000 ng/d treatment groups compared to the control(P<0.05).In conclusion,RFRP-3 appears to inhibit follicular development and steroid hormone secretion in mice by modulating the expression of apoplotic factors,oocyte secretion factors,and steroidogenic enzymes within the ovaries.This study provides a theoretical foundation for understanding the regulatory mechanisms of RFRP-3 on mammalian follicular development and steroid hormone synthesis.
