3种葱属植物的基因组大小及染色体倍性测定

【摘要】 为探索并建立一种适用于葱属植物的流式细胞术(FCM)检测体系,实现对实葶葱、分蘖洋葱和楼葱等3种葱属植物的染色体倍性和基因组大小的高效、精确测定,以嫩叶为材料,洋葱作为参照标准,对5种解离液进行了筛选和优化,并用根尖染色体压片法验证倍性检测结果。结果表明,改良LB01解离液解离效果最佳;测得实葶葱、分蘖洋葱、楼葱的基因组分别为13.74、15.40、17.96 Gb;实葶葱、分蘖洋葱均为二倍体,楼葱为三倍体,流式细胞术检测结果与根尖压片结果一致。

【Abstract】 In order to explore and establish a flow cytometry(FCM) detection system suitable for Allium species, and to achieve efficient and accurate determination of chromosome ploidy and genome size of 3 Allium species, such as Allium galanthum, Allium cepa L. var. aggregatum G. Don and Allium fistulosum L. var. viviparum Makino. In this study, young leaves were used as materials, and onion was used as the reference standard. Five kinds of lysate were screened and optimized, and the chromosome tablet method was used to verify the ploidy test results. The results showed that the improved LB01 dissociation solution had the best dissociation effect. The genomes of Allium galanthum Kar. & Kir., Allium cepa L. var. aggregatum G. Don and Allium fistulosum L. var. viviparum Makion were 13.74 Gb, 15.40 Gb and 17.96 Gb, respectively. The results of flow cytometry were consistent with that of root tip tablet.
