不同方式长期保藏黑木耳菌种性状的效果
Study on the traits of Auricularia heimuer strains preserved for long time by different methods
【摘要】 为研究不同保藏方法对黑木耳菌种性状的影响,以10株分别采用液氮保藏和4℃斜面移植法保藏20年的黑木耳菌种为供试材料,采用生长状态测定、栽培出耳、酯酶同工酶和ISSR分子标记等方法对不同保藏方式的长期保藏效果进行研究。结果表明,同一菌种在菌丝长势和生长速度方面,液氮保藏整体表现显著优于4℃斜面移植法保藏;出耳方面未见显著差异。不同保藏方式下的同一菌种的酯酶同工酶聚类分析图谱相似系数均为1,ISSR分子标记聚类分析中相似系数均高于0.90,说明不同保藏方式下同一菌种之间高度相似,但仍存在一定差异,可能是菌种长期4℃斜面保藏或者传代过程中出现了变化。研究结果对黑木耳菌种长期保存以及保持生产性状的稳定性具有重要意义。
【Abstract】 To study the effects of different preservation methods on the traits of Auricularia heimuer strains, 10 strains of A. heimuer that had been preserved for 20 years by liquid nitrogen preservation method and 4℃ slant transplantation method respectively were used as the test materials. Methods such as determination of growth status, cultivation for fruiting body production, esterase isoenzyme analysis, and ISSR molecular marker analysis were adopted to investigate the long-term preservation effects of different preservation methods. The results showed that for the same strain, in terms of mycelial growth vigor and growth rate, the overall performance of liquid nitrogen preservation was significantly better than that of the 4℃ slant transplantation method. There was no significant difference in the aspect of fruiting body production. For the same strain under different preservation methods, the clustering analysis of the esterase isoenzyme indicated was uniformly 1, and the similarity coefficients in the ISSR molecular marker clustering analysis were all higher than 0.90. This indicates that the same strain under different preservation methods is highly similar but there are still certain differences. It is likely that changes occurred during the long-term preservation of the strain by the 4 °C slant transplantation method or during the subculture process. This study is of great significance for the long-term preservation of A.heimuer strains and maintaining stability during the process of industrial production.
