超声协同维生素B诱导桦褐孔菌高产多糖
Ultrasound Combined with Vitamin B Synergistic Induced High-Yield Polysaccharides of Inonotus obliquus
【摘要】 本研究旨在通过超声波与外源因子协同诱导桦褐孔菌高产多糖,突破桦褐孔菌多糖产量低,不能满足应用需求的技术瓶颈。以桦褐孔菌胞外多糖为主要优化指标,筛选最佳超声诱导条件(菌龄5 d,超声温度40℃,超声时间15 min,超声功率50 W)与最佳外源因子以及添加质量浓度(VB260μg/mL),协同诱导桦褐孔菌高产多糖。超声与外源因子单一诱导后桦褐孔菌多糖含量分别为288.534 mg/150 mL和290.396 mg/150 mL,而协同诱导组多糖含量达302.375 mg/150 mL,均高于单一处理组。超声可显著促进营养物质进入细胞,使得桦褐孔菌多糖的产量高达244.149 mg/150 m L,外源因子的添加提高了糖代谢的相关酶活,添加60μg/mL VB2于培养基内,EPS产量可达231.679 mg/150 mL。相比单一处理方式而言,协同诱导可进一步提高细胞通透性、提高GALE酶(UDP-葡萄糖向UDP-半乳糖转化酶)、UDP-葡萄糖焦磷酸化酶等相关酶活性而激活部分糖醛酸通路,EPS含量提高了51.01%。同时,可通过改变协同诱导技术中的葡萄糖、甘露糖、半乳糖参数,优化桦褐孔菌多糖的DPPH自由基清除能力。
【Abstract】 A new technique combining ultrasonic and exogenous factors(cooperative induction technique, CIT) to induce Inonotus obliquus to high yield polysaccharide was proposed. The CIT could break through the technical bottleneck that the low yield of Inonotus obliquus polysaccharide could not meet the application requirements. The extracellular polysaccharide of Inonotus obliquus was used as the main optimization indicator, the optimal ultrasonic induction conditions(age of 5 d, ultrasonic temperature of 40 ℃, ultrasonic time of 15 min, and ultrasonic power of 50 W) and the optimal exogenous factors and addition concentration(VB260 μg/mL) to induce high polysaccharide production in Inonotus obliquus were determined. The polysaccharide contents of Inonotus obliquus were 288.534 mg/150 mL, and 290.396 mg/150 m L,respectively, while the polysaccharide content of the CIT group could be 302.375 mg/150 mL, which was higher than that of the single ultrasonic induction group and exogenous factors induction group. Ultrasonication significantly enhances nutrient uptake into cells, yielding a maximum production of Inonotus obliquus polysaccharides at 244.149 mg/150 m L.The addition of exogenous factors increased the activity of enzymes involved in carbohydrate metabolism; incorporating 60μg/mL vitamin B2 into the medium resulted in an extracellular polymeric substance(EPS) yield of 231.679 mg/150 m L.Compared to single treatments, synergistic induction further enhances cell permeability and activates enzymes such as GALE(UDP-glucose transaminase), UDP-glucose pyrophosphorylase, and other related enzymes, thereby activating portions of the uronic acid pathway. This resulted in a 51.01% increase in EPS content. Meanwhile, the DPPH radicalclearing ability of Inonotus obliquus polysaccharides could be optimized by changing the glucose, mannose, and galactose parameters in the synergistic induction technique.
