牡蛎肽与海参肽的活性功能比较
Comparison of the Active Functions of Oyster Peptides and Sea Cucumber Peptides
【摘要】 海参肽(SCP)和牡蛎肽(OP)在能量调控、激素水平和生殖功能等方面均有相似活性,然而二者的差异性仍未见研究报道。以海参肽和牡蛎肽为研究对象,评估二者在体内、外抗氧化活性、抗疲劳作用、激素调节能力的活性差异,并采用主成分分析(PCA)和支持向量机(SVM)方法挖掘区分二者的功能指标。结果显示,SCP和OP分子质量小于2 ku的肽分别占比95.94%和98.97%,主要氨基酸分别为甘氨酸和谷氨酸。SCP和OP均能有效清除OH-、DPPH和O2-自由基,且体外模拟消化不影响其效果。其中,OP的DPPH清除能力显著高于SCP,达到89.18%。体内研究结果表明,SCP组的总抗氧化能力、乳酸水平和尿素氮水平分别为14.36 U/mg prot,0.48 mmol/L,4.32 mmol/L,且与OP组比较均有统计学差异,然而丙二醛和超氧化物歧化酶无显著性差异。PCA和SVM分析显示,SCP和OP最主要的功效差异表现在激素调节方面:SCP对睾酮、促卵泡激素和促黄体生成素的调节能力强于OP。然而,OP在降低5型磷酸二酯酶水平方面更显著,比SCP组高1.58 ng/L。本研究比较了SCP和OP的生物活性差异,为活性肽在个性化预防和精准营养中的应用提供了理论参考。
【Abstract】 Sea cucumber peptides(SCP) and oyster peptides(OP) have similar activities in energy regulation, hormone levels, and reproductive function, but there have been no research reports on the differences between the two. This study aims to evaluate the differences in antioxidant activity, anti-fatigue effect, and hormone regulating ability between the SCP and OP in the experiment of in vitro and in vivo. Principal component analysis(PCA) and support vector machine(SVM) methods were also used to find discriminative functional indicators. The results showed that peptides with molecular weights less than 2 ku accounted for 95.94% and 98.97% in SCP and OP, respectively, and the major amino acids were glycine and glutamic acid. Both SCP and OP effectively scavenge OH-, DPPH, and O2-radicals, and their efficacy remains unaffected by simulated in vitro digestion. Notably, OP exhibite d significantly higher DPPH scavenging capacity than SCP, reaching 89.18%. In vivo studies revealed that the total antioxidant capacity, lactate levels, and blood urea nitrogen levels in the SCP group were 14.36 U/mg prot, 0.48 mmol/L, and 4.32 mmol/L, respectively, all showing statistically significant differences compared to the OP group. However, no significant differences were observed in malondialdehyde and superoxide dismutase levels. PCA and SVM analyses revealed that the primary efficacy difference between SCP and OP lies in hormonal regulation: SCP demonstrated stronger regulatory effects on testosterone, follicle-stimulating hormone, and luteinizing hormone than OP. Conversely, OP demonstrated a more pronounced reduction in phosphodiesterase type 5 levels, exceeding the SCP group by 1.58 ng/L. This study compared the differences in biological activity between SCP and OP, providing a theoretical basis for the application of active peptides in personalized prevention and precision nutrition.
