甘肃省某规模化奶牛场犊牛腹泻病原诊断及治疗分析

【摘要】 为了解甘肃省某规模化奶牛养殖场犊牛腹泻的病原携带情况,探究中药泻痢康口服液的治疗效果,使用5联犊牛腹泻病原微生物核酸检测试剂盒对该养殖场采集的10份犊牛腹泻粪便样本进行牛轮状病毒(Bovine Rotavirus, BRV)、牛冠状病毒(Bovine Coronavirus, BCoV)和产气荚膜梭菌(Clostridium perfringens,Cp)检测;对Cp核酸阳性粪便进行细菌分离培养、鉴定、遗传进化分析、药敏检测,并分析泻痢康口服液的治疗效果。结果表明:采集的犊牛腹泻粪便样本中Cp核酸阳性率为90%(9/10),BRV核酸阳性率为40%(4/10),BCoV核酸阳性率为30%(3/10)。Cp、BRV、BCoV的混合感染率为50%(5/10),确诊该病例为牛轮状病毒、冠状病毒和产气荚膜梭菌混合感染。细菌分离得到3株Cp菌株。16S rRNA遗传进化树分析发现,MGS分离株与巴基斯坦水牛源菌株(MUZ3-IOM-23)、伊拉克山羊源菌株(AKB38Duhok)具有较近的亲缘性,与其他菌株(VNF01、KM-1、CP1、BG01、4928STDY7387880)属于同一支。药敏试验结果显示,66.67%以上的Cp分离株对青霉素和庆大霉素耐药,对红霉素比较敏感。泻痢康口服液能够缓解犊牛的腹泻程度,缩短病程。综上所述,该养殖场犊牛腹泻主要是由Cp、BCoV和BRV混合感染引发,Cp分离株对青霉素和庆大霉素耐药率高,泻痢康口服液可用于治疗犊牛腹泻。本研究为该养殖场犊牛腹泻的病因诊断和防治提供了理论依据。

【Abstract】 To understand the pathogen carrying situation of a case of calf diarrhea in a large-scale dairy farm in Gansu Province, and to explore the therapeutic effect of the traditional Chinese medicine Xielikang oral liquid. In this study, 10 fecal samples of calf diarrhea collected from the farm were tested for bovine rotavirus(BRV), bovine coronavirus(BCoV) and Clostridium perfringens(Cp) using a 5-link calf diarrhea pathogenic microorganism nucleic acid detection kit; bacterial isolation and culture, identification, genetic evolution analysis, drug sensitivity test and Xielikang oral liquid treatment analysis were performed on Cp nucleic acid positive feces. The results showed that the positive rate of Cp nucleic acid in the collected fecal samples of calf diarrhea was 90%(9/10), the positive rate of BRV nucleic acid was 40%(4/10), and the positive rate of BCoV nucleic acid was 30%(3/10). The mixed infection rate of Cp, BRV and BCoV was 50%(5/10). The case was confirmed to be a mixed infection of bovine rotavirus, coronavirus and Clostridium perfringens. A total of 3 strains of Cp were isolated. Analysis of the 16S rRNA genetic evolutionary tree revealed that the MGS isolate was closely related to the Pakistani buffalo strain(MUZ3-IOM-23) and the Iraqi goat strain(AKB38 Duhok), and belonged to the same branch as other strains(VNF01, KM-1, CP1, BG01, 4928 STDY7387880). The results of drug sensitivity test showed that more than 66.67% of Cp isolates were resistant to penicillin and gentamicin, and were relatively sensitive to erythromycin. Xielikang oral liquid can effectively alleviate the degree of diarrhea in calves and shorten the course of the disease. In summary, the diarrhea of calves in this farm is mainly caused by mixed infection of Cp, BCoV and BRV. The three isolates are all Clostridium perfringens, with high resistance to penicillin and gentamicin. Xielikang oral liquid is a potential drug for the treatment of calf diarrhea, which provides a theoretical basis for the etiology diagnosis and prevention of calf diarrhea in this farm.
