基于气相色谱-质谱联用技术的雄激素性脱发代谢组学差异分析
Metabonomics difference analysis of androgenetic alopecia based on GC-MS
【摘要】 目的 研究分析雄激素性脱发(AGA)患者与健康人群的血清代谢差异。方法 选取2023年1月至2023年6月江阴市中医院收治的40例雄激素性脱发患者作为AGA组,同时选取40例健康体检者作为健康对照组。采用气相色谱-质谱联用(GC-MS)技术对两组试验人群的血清样本进行代谢组学研究,利用主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA)寻找差异性代谢物,通过菲恩(FIEHN)数据库和美国国家标准与技术研究院(NIST)数据库分析差异性代谢物,并获取相关代谢通路。结果 GC-MS代谢组学分析共鉴别出35种差异代谢物和3条关键代谢通路,AGA组与健康对照组存在显著代谢差异。结论 AGA患者存在半乳糖代谢、淀粉和蔗糖代谢、亚油酸代谢紊乱。
【Abstract】 Objective To investigate and analyze serum metabolic differences between patients with androgenetic alopecia(AGA) and healthy individuals. Methods 40 AGA patients admitted to Jiangyin Hospital of Chinese Medicine from January to June 2023 were enrolled as the AGA group, and 40 healthy individuals undergoing physical examinations were enrolled as the healthy control group. Serum samples from both groups underwent metabolomic profiling using gas chromatography-mass spectrometry(GC-MS). Principal component analysis(PCA) and orthogonal partial least squares-discriminant analysis(OPLS-DA) were employed to identify differential metabolites. Differential metabolites and pathway analysis were performed using the Fiehn(FIEHN)Library and National Institute of Standards and Technology(NIST) databases. Results GC-MS metabolomics identified 35 differential metabolites and 3 key metabolic pathways demonstrating substantial metabolic disparities between the AGA group and the healthy control group. Conclusion AGA patients exhibit dysregulation in galactose metabolism, starch and sucrose metabolism, and linoleic acid metabolism.
