糖尿病肾脏病患者氧化应激及炎症因子与HIF-1α、VEGF的相关性分析
Correlation analysis of oxidative stress and inflammatory factors with HIF-1α and VEGF in DKD patients
【摘要】 目的 探讨糖尿病肾脏病(DKD)患者氧化应激、炎症反应与缺氧诱导因子-1α(HIF-1α)、血管内皮生长因子(VEGF)的相关性。方法 选取符合纳入标准的DKDⅢ~Ⅳ期患者94例,根据DKD分期进行分组,其中DKDⅢ期组48例,DKDⅣ期组46例。选取46例单纯2型糖尿病(T2DM)患者作为对照组,检测超氧化物歧化酶(SOD)、同型半胱氨酸(Hcy)、白介素-6(IL-6)、超敏-C反应蛋白(hs-CRP)、肿瘤坏死因子-α(TNF-α)、HIF-1α、VEGF的水平,分析三组患者氧化应激及炎症因子与HIF-1α、VEGF的相关性。结果 血清Hcy、IL-6、hs-CRP、TNF-α、HIF-1α、VEGF水平在对照组、DKDⅢ期组、DKDⅣ期组依次升高,SOD水平依次降低(P <0.05)。血清Hcy、IL-6、hs-CRP、TNF-α水平与HIF-1α、VEGF水平呈正相关,SOD水平与HIF-1α、VEGF水平呈负相关(P <0.05)。结论 DKD患者HIF-1α、VEGF水平随DKD分期进展而升高,且与炎症反应及氧化应激严重程度存在一定的相关性。
【Abstract】 Objective To investigate the correlation between oxidative stress,inflammatory response and HIF-1α,VEGF in diabetic kidney disease(DKD) patients.Methods A total of 94 patients with DKD stage Ⅲ~Ⅳ who met the inclusion criteria were enrolled, grouping according to DKD staging, 48 patients in the DKD stage Ⅲ group and 46 patients in the DKD stage Ⅳ group. A total of 46 patients with simple T2DM were selected as the control group.The levels of superoxide dismutase(SOD),homocysteine(Hcy), interleukin-6(IL-6),hypersensitivity-C-reactive protein(hs-CRP),tumor necrosis factor-α(TNF-α),hypoxia-inducible factor-1α(HIF-1α) and vascular endothelial growth factor(VEGF) were measured,to analyze the correlation of oxidative stress and inflammatory factors with HIF-1α and VEGF in the three groups of patients. Results Hcy, IL-6, hs-CRP, TNF-α, HIF-1α and VEGF increased in the control group, DKD stage Ⅲ group, and DKD stage Ⅳ group in order, and SOD decreased in the control group, DKD stage Ⅲ group, and DKD stage Ⅳ group in order(P < 0.05).Hcy, IL-6, hs-CRP, and TNF-α were positively correlated with HIF-1α and VEGF levels, while SOD was negatively correlated with HIF-1α and VEGF levels. Conclusion The levels of HIF-1α and VEGF in DKD patients increased with the progression of DKD stage, and there was a certain correlation with the severity of inflammatory response and oxidative stress.
