异甘草酸镁联合肝细胞生长因子对大鼠肝纤维化的影响
Effect of magnesium isoglycyrrhiza combined with hepatocyte growth factor on hepatic fibrosis in rats
【摘要】 目的 观察异甘草酸镁(Mg IG)联合肝细胞生长因子(HGF)对大鼠肝纤维化(HF)的治疗作用。方法 采用皮下注射40%四氯化碳(CCl4)-花生油溶液制备HF模型。试验分为HF模型组、HGF治疗组、Mg IG高剂量组、HGF+Mg IG低剂量组、HGF+Mg IG高剂量组及正常组。5周后检测各组肝功能指标[天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、γ-谷氨酰转移酶(GGT)、碱性磷酸酶(ALP)]、HF指标[层黏连蛋白(LN)、透明质酸(HA)、Ⅲ型前胶原(PCⅢ)、IV型胶原(ColⅣ)],以及炎症指标[白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、转化生长因子-β1(TGF-β1)];结合肝组织病理切片观察各组炎性反应及HF程度。结果 造模5周后,与正常组比较,CCl4组AST、ALT、ALP、GGT水平均显著升高(P<0.05)。给药5周后,与HF模型组比较,HGF+Mg IG联合用药组血清AST、ALT、ALP、GGT、HA、PCⅢ、ColⅣ、LN、IL-6、TNF-α、TGF-β1水平均显著降低(P<0.05),且和正常组相比,差异无统计学意义(P>0.05);病理切片观察到HGF+Mg IG低剂量组炎症消失,纤维化基本消失,HGF+Mg IG高剂量组肝组织恢复正常,表明HGF和Mg IG联合用药对大鼠HF的治疗效果更明显,显著优于单一用药。结论 HGF和Mg IG联合用药能显著减轻CCl4导致的HF。
【Abstract】 Objective To observe the therapeutic effect of magnesium isoglycyrrhiza (Mg IG) combined with hepatocyte growth factor(HGF) on hepatic fibrosis (HF) in rats.Methods A rat model of HF induced by carbon tetrachloride (CCl4) was used.It was divided into HF model group,HGF group,Mg IG treatment group,low-dose HGF+Mg IG group,high-dose HGF+Mg IG group and normal group.After 5weeks of treatment with the drugs,liver function indicators[(aspartate aminotransferase (AST),alanine aminotransferase (ALT),alkaline phosphatase (ALP),γ-glutamyl transferase (GGT)]and HF indicators[hyaluronic acid (HA),type III procollagen (PCⅢ),laminin (LN),type IV collagen (Col-IV)]as well as inflammatory indicators[interleukin-6 (IL-6),tumor necrosis factor-α (TNF-α),transforming growth factor-β1 (TGF-β1)]were detected in each group.The inflammatory response and the degree of HF in each group were observed by liver tissue pathological sections.Results After 5 weeks of moding,compared with the normal group,the levels of AST,ALT,ALP,GGT in the CCl4 group were significantly increased (P<0.05).After 5 weeks of treatment,compared with the HF model group,the combined treatment of HGF and Mg IG significantly reduce the levels of serum AST,ALT,ALP,GGT,HA,PCⅢ,ColⅣ,LN,IL-6,TNF-α,and TGF-β1 (P<0.05),there was no significant difference compared with the normal group (P>0.05).Pathological observation showed that in the low-dose HGF+Mg IG group,the inflammation disappeared and the fibrosis almost completely disappeared;in the high-dose HGF+Mg IG group,the liver tissue returned to normal and was comparable to the normal group,which was showed that the combination of HGF and Mg IG was more effective in rat HF,significantly better than the single-drug treatment.Conclusion The combined treatment of HGF and Mg IG can significantly alleviate HF induced by CCl4.
