深绿木霉T2菌株土壤定殖条件及其对苜蓿生长的影响
Soil Colonization Conditions of Trichoderma atroviride T2Strain and Its Effects on Alfalfa Growth
【摘要】 为探究深绿木霉T2菌株土壤定殖条件及其对苜蓿生长的影响,本研究采用土壤浇灌法研究不同浓度孢子悬浮液、土壤湿度及温度对T2菌株定殖的影响,并用筛选出的最佳浓度T2菌株孢子悬浮液浇灌处理土壤,分析其对苜蓿种子萌发及幼苗生长的影响。结果表明:T2菌株土壤定殖的最佳初始孢子浓度、土壤湿度、环境温度分别为1×10~7个/g、70%、30℃,在该培养条件下培养28 d后T2菌株孢子在土壤中定殖量达到最大,为5.95×10~7 cfu/g。T2菌株土壤定殖对苜蓿种子萌发和盆栽幼苗生长有促进作用,且促生效果明显。当土壤中初始T2菌株孢子浓度为1×10~7个/g时,T2菌株浇灌处理苜蓿种子发芽第3 d的萌发率和芽长的促生率分别为47.37%和37.50%;分别在处理第21、21、35 d时对盆栽幼苗的叶面积、株高、根长促生效果最好,促生率可达71.92%、72.08%、88.43%。
【Abstract】 In order to investigate the soil colonization conditions of Trichoderma atroviride T2 strain and its effects on alfalfa growth,soil watering method was used to determine the effects of different concen‐trations of T2 strain spore suspension,soil moisture and temperature on its colonization.Alfalfa seeds and potted seedlings were inoculated with the spore suspension of the T2 strain at the optimal concentration for soil colonization.This study aims to evaluate the effects on alfalfa seed germination and seedling growth.The results showed that the initial spore concentration,soil moisture and temperature conducive to the colo‐nization of T2 strain were 1×10~7 spores/g,70%and 30℃,respectively.After 28 days,the colonization amount of T2 strain spores in soil reached the maximum,which was 5.95×10~7 cfu/g.The soil coloniza‐tion of T2 strain can promote the germination of alfalfa seeds and the growth of potted seedling,and the growth promotion effect was obvious.When the initial spore concentration of T2 strain in soil was 1×10~7spores/g,the treatment of T2 strain on the 3rd day had the best effect on seed germination growth,the growth rate of germination rate and bud length were 47.37%and 37.50%,respectively.At 21 d,21 d and35 d after T2 treatment,the growth promotion effect on leaf area,plant height and root length of potted seedlings were the best,and the growth promotion rate reach 71.92%,72.08%and 88.43%,respec‐tively.
