猫爪草多糖对血管内皮细胞缺氧-复氧损伤的保护作用
Protective effect of polysaccharide of Radix Ranunculi Ternati on hypoxia-reoxygenation injury of vascular endothelial cells
【摘要】 目的 探讨猫爪草多糖(PRT)对血管内皮细胞缺氧-复氧(H-R)损伤的保护作用。方法 采用水提醇沉法及Sevag法从猫抓草中提取PRT,培养人脐静脉内皮细胞株EA·hy926并进行处理。EA·hy926细胞分别加入PRT 50、100、200、400和800mg/L,孵育24h后测定PRT对细胞的毒性作用。随后EA·hy926细胞借助低氧/厌氧工作站并结合酸性缺氧液建立H-R模型(H-R组);EA·hy926细胞分别加入PRT 50、100、200、400和800mg/L孵育12h,随后建立H-R模型;对照组细胞进行常规培养。采用CCK-8法检测细胞存活率,流式细胞术检测细胞活性氧水平,细胞划痕实验检测细胞迁移率。结果 与对照组比较,H-R组细胞存活率及迁移率降低,活性氧平均荧光强度升高(P<0.05)。与H-R组比较,PRT 200mg/L+H-R组、PRT 400mg/L+H-R组及PRT 800mg/L+H-R组的细胞存活率增加,PRT 400mg/L+H-R组、PRT 800mg/L+H-R组的细胞迁移率升高(P<0.05)。结论 PRT对血管内皮细胞H-R损伤有保护作用,能够增加细胞存活及迁移。
【Abstract】 Objective To investigate the protective effect of polysaccharide of Radix Ranunculi Ternati(PRT) on hypoxia-reoxygenation(H-R) injury of vascular endothelial cells.Methods PRT was extracted from Radix Ranunculi Ternati by water extract-alcohol precipitation and Sevag method.Human umbilical vein endothelial cell line EA·hy926 cells were cultured.EA·hy926 cells were added with PRT 50,100,200,400 and 800mg/L,respectively, and incubated for 24 hours to determine the toxic effects of PRT on the cells.The H-R model of EA·hy926 cells was established by hypoxic/anaerobic workstation combined with acid anoxic solution.EA·hy926 cells were added with PRT 50,100,200,400 and 800 mg/L for 12 hours before H-R model was established.The normal cells were cultured as the controls.The cell survival rate was measured by CCK-8 method, reactive oxygen species level was measured by flow cytometry, and cell migration rate was measured by cell scratch assay.Results Compared with control group, the survival rate and migration rate were decreased, and the mean fluorescence intensity of reactive oxygen species was increased in H-R group(P<0.05).Compared with H-R group, the survival rate was increased in PRT 200mg/L+H-R group, PRT 400mg/L+H-R group and PRT 800mg/L+H-R group(P<0.05),migration rate was increased in PRT 400mg/L+H-R group and PRT 800mg/L+H-R group(P<0.05).Conclusion PRT has protective effect on H-R injury of vascular endothelial cells, and could increase cell survival and migration.
