“密蒙花-夏枯草-陈皮-玫瑰花”药组治疗睑板腺功能障碍性干眼的网络药理学研究

【摘要】 目的 基于网络药理学探讨“密蒙花-夏枯草-陈皮-玫瑰花”药组治疗睑板腺功能障碍性干眼的作用机制。方法 利用中药系统药理学数据库与分析平台(TCMSP)和SwissTargetPredicton数据库筛选出药组的有效活性成分及作用靶点;应用人类基因数据库(GeneCards)和人类孟德尔遗传数据库(OMIM)筛选出与疾病相关的靶点;使用微生信在线工具绘制韦恩图,得到中药药组治疗疾病的交集靶点;使用Cytoscape 3.10.0软件构建“中药-成分-靶点-疾病”网络图筛选出关键成分,再利用STRING数据库构建蛋白互作网络筛选出核心靶点;最后通过David数据库和微生信绘图工具对交集靶点进行GO分析和KEGG分析。结果 中药药组中共挖掘出治疗疾病的有效活性成分21种,交集靶点103个,核心靶点21个。药物有效成分主要为维生素E、飞燕草素、槲皮素等,核心靶点主要涉及丝氨酸/苏氨酸激酶1(AKT1)、肿瘤坏死因子(TNF)、表皮生长因子受体(EGFR)、雌激素受体1(ESR1)等,GO生物学过程主要包括肽基-酪氨酸磷酸化、蛋白质自磷酸化、MAPK级联激活等,KEGG主要包括PI3K-Akt通路、MAPK通路等。结论 “密蒙花-夏枯草-陈皮-玫瑰花”药组可能通过PI3K-Akt通路、HIF-1通路、MAPK通路,作用于AKT1、TNF、EGFR、SRC、ESR1、BCL2、HIF1A等靶点,发挥抗炎、抗氧化、抗细胞凋亡等功能,起到治疗睑板腺功能障碍性干眼的作用。

【Abstract】 Objective To investigate the mechanism of the “Mimenghua(Buddlejae Flos)-Xiakucao(Prunellae Spica)-Chenpi(Citrus Reticulata)-Meiguihua(Rose Rugosae Flos)” combination in treating meibomian gland dysfunction-related dry eye disease based on network pharmacology.Methods All the effective active ingredients and targets of action of the TCM pharmacome were screened out by using Traditional Chinese Medicine Systems Pharmacology(TCMSP) and SwissTargetPredicton database.Targets related to this disease were screened through Human Genes(GeneCards) and Online Mendelian Inheritance in Man(OMIM).The intersecting targets of the TCM pharmacome in treating this disease were obtained by using WeiShengXin software to draw Venn diagram.A network diagram of “drug-active component-target-disease” was drawn by using Cytoscape 3.10.0 software and key components were selected. The STRING database was then used to construct a protein-protein interaction(PPI) network to screen out the core targets. Finally, Gene Ontology(GO) analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG) analysis of the intersecting targets were performed by the David database and WeiShengXin mapping tools.Results A total of 21 effective active ingredients for the treatment of this disease, 103 intersecting targets and 21 core targets have been excavated from the TCM pharmacological group.The active ingredients of Chinese medicine are mainly vitamin-e, delphinidin, quercetin, etc.Core targets mainly involve AKT1, TNF, EGFR, ESR1, etc.GO biological processes mainly include peptidyl-tyrosine phosphorylation, protein autophosphorylation, positive regulation of MAPK cascade, etc.KEGG mainly includes the PI3K-Akt pathway, MAPK pathway, etc.Conclusion “Mimenghua-Xiakucao-Chenpi-Meiguihua” combination may act on AKT1,TNF,EGFR,SRC,ESR1,BCL2,HIF1A and other targets through PI3K-Akt, HIF-1,MAPK signaling pathway, which exerts anti-inflammatory, antioxidant and anti-apoptotic functions, and plays a role in treating meibomian gland dysfunction-related dry eye disease.
