山东地区猪流行性腹泻病毒分子流行病学调查及流行毒株的分离鉴定
Molecular epidemiological investigation of porcine epidemic diarrhea virus and isolation and identification of epidemic strains in the Shandong area
【摘要】 猪流行性腹泻病毒(PEDV)感染可引起仔猪急性腹泻和脱水,死亡率高。为了明确山东地区PEDV的流行趋势与遗传进化情况,本研究于2022—2023年对山东省的13个地级市进行了流行病学调查。通过对PEDV N基因的RT-PCR检测,结果在228份疑似腹泻样本中,PEDV阳性率达48.68%;同时,在PEDV感染的样本中,发现PEDV与猪德尔塔冠状病毒(PDCoV)、猪轮状病毒(PoRV)共感染的现象。对PEDV阳性样本进行S1基因的扩增测序,通过MEGA6软件对PEDV阳性样本进行S1基因的遗传进化分析,发现扩增的4株流行毒株属于GⅡc型,1株流行毒株属于GⅡa型。进一步对流行毒株进行分离,通过PEDV N基因的检测和间接免疫荧光染色,证实成功分离到1株PEDV(命名为SDQD20230628)。对分离株进行S基因的遗传进化分析,发现分离株S基因与GⅡc亚型毒株同源性较近。本研究发现PEDV仍是引起山东地区猪场腹泻猪主要病毒,且呈现出GⅡc亚型毒株趋于流行的趋势,研究结果丰富了山东地区PEDV分子流行病学数据,也为PEDV感染的防控提供了理论依据。
【Abstract】 Porcine epidemic diarrhea virus(PEDV) infection causes acute diarrhea and dehydration in piglets, with a high mortality rate, posing a serious threat to the healthy development of the swine industry in China. In order to clarify the epidemic trend and genetic evolution of PEDV in Shandong Province, we conducted an epidemiological survey in 13 prefecture-level cities in Shandong Province from 2022 to 2023. The detection of the N gene of PEDV showed a positive rate of PEDV in 228 suspected diarrhea samples to be 48.68%. Meanwhile, the co-infection of PEDV with porcine deltacoronavirus(PDCoV) and porcine rotavirus(PoRV) was found in the PEDV-positive samples. Subsequently, the S1 gene of PEDV-positive samples was amplified and sequenced, and the genetic evolution analysis of PEDV-positive samples was conducted using MEGA6 software, which showed that four prevalent strains belonged to the GⅡc type and one strain belonged to the G Ⅱ a subtype. We further isolated the prevalent strains and confirmed the successful isolation of a PEDV strain( named SDQD20230628) through detection of the N gene and indirect immunofluorescence staining. The S gene genetic evolution analysis of the isolated strain showed that the isolated strain had a close homology with the GⅡc subtype. It was found that PEDV was still the main virus causing diarrhea in pig farms in Shandong Province, and a trend was emerging that the GⅡc subtype strains were getting prevalent. This study not only enriched the molecular epidemiological data of PEDV in Shandong Province, but also provided important theoretical support for prevention and control of PEDV.
