番茄红素对脂多糖诱导的滑膜细胞炎症反应抑制作用的研究
Exploring the inhibitory effects of lycopene on lipopolysaccharide induced inflammatory response in synovial cells
【摘要】 目的 探讨番茄红素是否对脂多糖(lipopolysaccharide, LPS)诱导的滑膜细胞炎症反应有抑制作用。方法 体外培养的人滑膜成纤维细胞,分组为对照组(不干预);模型组(1μg/ml的LPS处理);低、中、高浓度组(0.1、0.5和2.5μmol/L番茄红素单独处理及干预模型组),处理时间均为24 h。采用CCK-8法检测细胞活性;ELISA法检测细胞上清液中炎性因子IL-1β和TNF-α水平。结果 与对照组比较,模型组、0.5和2.5μmol/L番茄红素单独处理组细胞活性均呈明显增加,差异有统计学意义(F=17.99,F=6.13,F=12.13,P<0.01或P<0.05);与模型组比较,2.5μmol/L番茄红素干预组细胞活性明显增加,差异有统计学意义(F=9.88,P<0.01)。与对照组比较,模型组细胞炎性因子IL-1β和TNF-α的表达明显增高,差异有统计学意义(F=41.97,F=82.48,P<0.01)。与模型组比较,0.1、0.5和2.5μmol/L的番茄红素干预组IL-1β和TNF-α的浓度明显降低,差异有统计学意义(F=19.25,F=28.52,F=31.44;F=28.12,F=43.85,F=59.37,P<0.01),且有明显的剂量-反应关系。结论 番茄红素对LPS诱导的滑膜细胞炎症反应有明显的干预作用。
【Abstract】 Objective Exploring whether lycopene has an inhibitory effect on lipopolysaccharide-induced inflammatory response in synovial cells.Methods Human synovial fibroblasts were cultured in following conditions: control(without intervention); model(treated with 1 μg/ml LPS); low, medium, and high-dose(0.1, 0.5, 2.5 μmol/L lycopene separately and both lycopene + 1 μg/ml LPS as well). All cells were treated for 24 hours, respectively.The cell viability was detected by CCK-8 kit.The levels of inflammatory factors IL-1β and TNF-α in the supernatant were detected by ELISA kit.Results Compared with the control group, cell-viability of the model group, 0.5 and 2.5 μmol/L lycopene treatment groups showed a significant increase, and the difference had statistically significant(F=17.99,F=6.13,F=12.13,P<0.01 or P<0.05). Compared with the model group, LPS combined with 2.5 μmol/L lycopene treatment showed a significant increase in cell viability, and the difference had statistically significant(F=9.88,P<0.01).Compared with the control group, the expression of inflammatory cytokines IL-1 β and TNF-α in the model group was significantly increased, and the difference was statistically significant(F=41.97,F=82.48,P<0.01).The concentrations of IL-1β and TNF-α decreased in all lycopene treatment(0.1, 0.5 and 2.5 μmol/L) plus LPS exposure condition when compared with the model group, and the difference were statistically significant(F=19.25,F=28.52,F=31.44;F=28.12,F=43.85,F=59.37,P<0.01), with a certain dose-response relationship as well.Conclusion Lycopene has a significant protective effect on LPS induced synovial cell inflammation response.
