ABA对大蒜愈伤组织中大蒜素含量的提升以及利用响应面法对大蒜素提取条件的优化

【摘要】 为优化愈伤组织生产大蒜素的体系并提高大蒜素的含量,以宝坻大蒜“六瓣红”的贮藏叶为外植体,在愈伤组织的诱导和增殖培养基中添加不同质量浓度的外源脱落酸(ABA),寻求诱导大蒜素生成的最佳质量浓度.对生产工艺中的5个因素(料液比、浸提时间、乙醇体积分数、酶解温度、酶解时间)进行单因素分析,根据响应值大小,筛选出影响较大的因素.在此基础上,通过响应面回归分析进一步优化大蒜素的提取体系.结果显示:(1)ABA质量浓度为0.10 mg/L时愈伤组织的诱导率最高,为96.67%,且继一代愈伤组织中大蒜素的含量最高,为10.09μg/g;(2)通过单因素分析筛选出3个影响较大的因素,即料液比、浸提时间和乙醇体积分数;(3)通过响应面回归分析获得了最优体系,即料液比为1∶6、浸提时间为1.6 h、乙醇体积分数为80%,大蒜素提取含量预测值为18.55μg/g;(4)利用最优体系提取愈伤组织大蒜素,验证值为18.35μg/g,预测值与验证值基本相符,与原方法相比大蒜素含量提高了185.64%.

【Abstract】 To optimize the system for allicin production in callus and enhance the allicin content,different mass concentrations of exogenous abscisic acid(ABA)were supplemented into the callus induction and proliferation media to identify the optimal mass concentration for inducing allicin synthesis,using the storage leaves of Baodi garlic “Liu Ban Hong” as explants. A single-factor analysis was conducted on five parameters in the production process(solid-liquid ratio,extraction time,ethanol volume fraction,enzymatic hydrolysis temperature and enzymatic hydrolysis time),the factors with greater influence were screened according to the influence of different factors on the response value. On this basis,the allicin extraction process was further optimized by response surface regression analysis. The results showed that:(1) The highest induction rate of callus tissue was 96.67% at ABA mass concentration of 0.10 mg/L,and the highest allicin content in the subcultured callus was 10.09 μg/g.(2) Three influential factors were identified through single factor analysis,namely the solid-liquid ratio,extraction time and ethanol volume fraction.(3) The optimal system was obtained through response surface regression analysis,with a solid-liquid ratio of 1 ∶ 6,extraction time of 1.6 h,ethanol volume fraction of 80%,and the predicted content of allicin extraction was 18.55 μg/g.(4) The callus allicin was extracted using the optimal system,and the verification value was 18.35 μg/g. The predicted value was basically consistent with the verification value,and the allicin content increased by 185.64% compared with the original method.
