加减藿朴夏苓汤对厌食模型大鼠脑肠肽及肠道菌群的影响

【摘要】 目的 探究加减藿朴夏苓汤对厌食大鼠脑肠肽及肠道菌群的影响。方法 将40只SD大鼠,随机分为空白对照组(10只),建模组(30只),采用病因模拟法造模成功后,将建模组随机分为模型组、阳性对照组、治疗组,每组10只,治疗组、阳性对照组予相应药物,空白对照组和模型组予等量生理盐水,连续灌胃14d,监测大鼠24h摄食量、体质量,用Western Blot法检测大鼠下丘脑神经肽Y(NPY)、胆囊收缩素8(CCK8)蛋白的表达,16s rDNA测序分析大鼠肠道微生物的变化。结果 加减藿朴夏苓汤可明显增加厌食模型大鼠的24h摄食量和体质量,升高NPY、降低CCK8蛋白的表达。与空白对照组比较,模型组放线菌门(Actinobacteria)及罗姆布茨菌属(Romboutsia)等8种菌属相对丰度降低,变形菌门(Proteobacteria)及韦荣氏球菌属(Veillonella)相对丰度升高,与模型组比较,治疗组拟杆菌门(Bacteroidetes)及Papillibacter、Prevotellaceae_Ga6A1_group相对丰度降低,厚壁菌门(Firmicutes)、Actinobacteria及罗氏菌属(Roseburia)等10种菌属相对丰度升高。结论 加减藿朴夏苓汤可增加厌食模型大鼠的摄食量和体质量,其作用机制可能与调节脑肠肽蛋白的表达及肠道菌群作用有关。

【Abstract】 Objective To investigate the effects of modified Huopu Xiangling Decoction(藿朴夏苓汤, HPXLD) on brain-gut peptides and intestinal microbiota in anorexia model rats.Methods Forty SD rats were randomly divided into the blank control group(10 rats) and the modeling group(30 rats). After successful model establishment using the etiological modeling method, the modeling group was randomly divided into the model group, positive control group, and the treatment group, with 10 rats in each group. The treatment group and the positive control group received corresponding drugs, while the blank control group and the model group were given equal volumes of physiological saline. All groups were administered by gavage for 14 consecutive days, during which the 24-hour food intake and body weight were monitored. Western blot(WB) analysis was performed to detect the expression of hypothalamic neuropeptide Y(NPY) and cholecystokinin-8(CCK-8) proteins. 16S rRNA gene sequencing was employed to analyze changes in intestinal microbiota. Results Modified HPXLD significantly increased the 24-hour food intake and body weight of anorexia model rats, increased NPY protein expression, and decreased CCK-8 protein expression. Compared with the blank control group, the model group exhibited decreased relative abundance of Actinobacteria and Romboutsia(among eight other genera), and increased relative abundance of Proteobacteria and Veillonella. Compared with the model group, the treatment group showed decreased relative abundance of Bacteroidetes, Papillibacter, and Prevotellaceae_Ga6A1_group, and increased relative abundance of Firmicutes, Actinobacteria, and Roseburia(among ten other genera). Conclusion Modified HPXLD increased food intake and body weight in anorexia model rats, and its mechanism may be associated with the regulation of brain-gut peptide expression and modulation of intestinal microbiota.
