【摘要】 乳酸乳球菌(Lactococcus lactis)作为一种食品级的安全菌株,是表达外源蛋白的优良宿主。基于乳链菌肽自我诱导表达机制改造获得的乳酸乳球菌NICE表达系统具有诱导效率高、自身分泌蛋白少、无胞外蛋白酶等特点,已被广泛用于表达各类外源蛋白。通过设计不同的表达元件,实现外源蛋白在胞内、胞外或细胞表面锚定表达,从而发挥蛋白质作为抗原、抗菌肽或作为酶制剂的生物学功能。综述了NICE表达系统的构建和诱导表达机制,总结了重组乳酸乳球菌在食品工业和黏膜疫苗等领域的研究现状,为乳酸乳球菌NICE表达系统未来的应用方向提供参考。

【Abstract】 Lactococcus lactis as a food-grade safe strain, is an excellent host for foreign protein expression. Based on nisin self-inducing expression mechanism, L. lactis NICE expression system is obtained and has been widely used for expression of various foreign proteins with characteristics of high induction efficiency, less self-secreted proteins, and no extracellular protease. By designing different expression elements, the exogenous proteins can be expressed intracellularly, extracellularly or on the cell surface, so as to play its role. This review introduces the construction and inducible expression mechanism of the NICE expression system, and summarizes the research status of recombinant L. lactis in the fields of food industry, medicine and health, and provides a reference for the future application of the L. lactis NICE expression system.
