基于桔梗皂苷D检测的HPLC-CAD试验设计
Experimental Design of HPLC-CAD Based on Detection of Platycodon Saponin D
【摘要】 建立测定桔梗原料中桔梗皂苷D含量的高效液相色谱-电雾式检测器(HPLC-CAD)仪器分析试验设计。基于Thermo ScientificTMCoronaTMVeoTMRS CAD检测器,以中药原料桔梗为例,建立桔梗皂苷D的高效液相色谱法,并对方法的可行性、可靠性和适用性进行分析。结果表明,桔梗皂苷D在0.5~50μg/mL之间进样量浓度与峰面积对数呈良好线性关系A=0.039 7C+0.065 4 (R2=0.992 6),样品稀释液浓度为1.70μg/mL,平均加样回收率为112.8%(SRSD=1.9%)。试验设计可靠、准确,具有良好的重复性,为桔梗原料的质量标准提升奠定基础,并能激发学生的科研兴趣,提高学生的科研素养和创新能力。
【Abstract】 Establish an experimental design for the determination of platycodon saponin D in platycodonplatycodon raw materials by high performance liquid chromatography with charged aerosol detector(HPLC-CAD). A method is established based on Thermo ScientificTM CoronaTM VeoTM RS CAD detector and high performance liquid chromatography(HPLC) for the determination of platycodonplatycodon saponin D from platycodongrandiflorus. The feasibility, reliability and applicability of the method are analyzed. The results show that the concentration of platycodon D in the range of 0.5-50 μg/mL shows a good linear relationship with the peak area, A=0.039 7C+0.065 4(R2=0.992 6), and the concentration of sample diluent is 1.70μg/m L. The average recovery is 112.8%(SRSD=1.9%). The experimental design is reliable, accurate and repeatable. It can not only lay a foundation for the improvement of the quality standard of platycodon grandidon raw materials, but also stimulate students’ interest in scientific research and improve their scientific research literacy and innovation ability.
