熊胆粉调节慢性不可预知应激模型小鼠肠-脑轴抗抑郁作用和机制研究
Study on antidepressant effect and mechanism of bear bile powder in regulating gut-brain axis in mice with chronic unpredictable stress model
【摘要】 目的:探索熊胆粉(BBP)对慢性不可预知应激(CUMS)模型小鼠的抗抑郁效果及作用机制。方法:将小鼠分为对照组、模型组、阳性药组和不同剂量BBP组,CUMS刺激3周后,建立慢性不可预知应激模型,灌胃药物干预21 d,进行行为学测试、海马神经递质检测、粪菌16S rDNA测序和粪菌移植实验。结果:BBP显著提升小鼠糖水偏好率(P<0.001),减少游泳和悬尾不动时间(P<0.05,P<0.01),增加海马5-羟色胺(5-HT)和5-羟吲哚乙酸(5-HIAA)含量(P<0.05),改善肠道菌群多样性,提高毛螺菌科(Lachnospiraceae)丰度,降低产碱杆菌科(Alcaligenaceae)丰度。并且BBP粪菌移植能明显减轻CUMS小鼠抑郁症状,并提升海马5-HT水平。结论:BBP通过调节肠-脑轴发挥抗抑郁作用。
【Abstract】 Objective: To explore the antidepressant effect and mechanism of bear bile powder(BBP) in chronic unpredictable mild stress(CUMS) mice. Methods: All mice were divided into control, model, positive drug, and different doses of BBP groups. After 3 weeks of CUMS stimulation, the mice were orally administered with drugs for 21 days for behavioral tests, hippocampal neurotransmitter detection, fecal 16S rDNA sequencing, and fecal microbiota transplantation experiment. Results: BBP significantly increased the sugar water preference rate of mice(P<0.001), reduced swimming and tail suspension time(P<0.05, P<0.01), increased hippocampal 5-hydroxytry(5-HT) and 5-hydroxyindole-3-acetic acid(5-HIAA) content(P<0.05), improved intestinal microbiota diversity, increased the proportion of Lachnospiraceae microbiota, and decreased the proportion of Alcaligenaceae microbiota. BBP fecal microbiota transplantation significantly alleviated depressive symptoms in CUMS mice and elevated hippocampal 5-HT levels. Conclusion: BBP exerted antidepressant effects by regulating gut-brain axis in mice.
