厦门地区A组轮状病毒VP4基因序列特征
Sequence characteristics of VP4 gene of group A rotavirus in Xiamen
【摘要】 目的 了解厦门地区P型A组轮状病毒VP4基因序列和遗传变异特征,及其与国内外VP4基因的差异。方法 收集2021年1—12月厦门市儿童医院100例<10岁腹泻患儿轮状病毒检测阳性的粪便样本。采用巢式聚合酶链反应(PCR)进行A组轮状病毒G基因型和P基因型分型。用一代测序技术进行A组轮状病毒VP4基因全长序列测序,用DNA Star、MEGA等生物学软件对测序后获得的基因序列进行系统进化分析和氨基酸序列比对。结果 100例腹泻患儿粪便样本G基因型主要流行株为G9型(88株),P基因型主要流行株为P[8]型(87株),G/P组合基因型主要流行株是G9P[8]型(80株)。进化树分析结果显示,厦门地方株(Amoy)与国内外其他地方株在进化树内的关联性存在不同程度的差异。氨基酸比对分析结果显示,厦门地方株(Amoy)与国内外不同地方株的氨基酸一级结构存在不同程度的变异。结论 A组轮状病毒VP4基因全长可显示毒株在中国具有快速适应和变异的能力。建议对轮状病毒的遗传变异进行持续监测。
【Abstract】 Objective To understand the VP4 gene sequence and genetic variation characteristics of type P rotavirus in Xiamen,as well as its differences from those of other regions at home and abroad. Methods From January to December 2021,100 fecal samples from children <10 years old with positive rotavirus determination result at the Children’s Hospital of Xiamen were collected. Nucleic acid polymerase chain reaction(PCR) was used for the genotyping of rotavirus G genotype and P genotype. The full-length VP4 gene sequence of rotavirus A was determined by first-generation sequencing technology. The sequences were analyzed and compared using biological softwares such as DNA Star and MEGA. Results The main prevalent strain of G gene in the 100 diarrhea samples was G9 type(88 strains),the main prevalent strain of P genotype was P[8] type(87 strains),and the main prevalent combination of G/P gene was G9P[8] type(80 strains). The evolutionary tree analysis showed that the local strains in Xiamen(Amoy) had different degrees of differences in the evolutionary tree compared with other local strains at home and abroad. The amino acid comparison analysis showed that the amino acid primary structure of the local strain in Xiamen(Amoy) had different degrees of variations compared with different local strains at home and abroad. Conclusions The full-length VP4 gene of rotavirus A can show that the strain has the ability to adapt and mutate rapidly in China. It is recommended to continuously monitor the genetic variation of rotavirus.
