茯苓多糖对上海白猪精液冷冻保存及精子DNA甲基化的影响

【摘要】 为研究茯苓多糖(Poria cocos mushroom polysaccharides,PCP)在上海白猪精液冷冻保存中的作用,采集8头上海白猪24份精液,在精液冷冻液中添加不同浓度的PCP,测定冷冻-解冻后精液质量和抗氧化活性,以及冷冻保存后精子DNA甲基化水平。结果表明:与未添加PCP组相比,600μg∕mL PCP添加组精子活力最高(P<0.05),600μg∕mL PCP添加组精子活率最高(P<0.05),而600μg∕mL和900μg∕mL PCP添加组的精子顶体完整率、质膜完整率、线粒体活性、活性氧(ROS)和丙二醛(MDA)含量较高。此外,600μg∕mL PCP添加组精液中的超氧化歧化酶(SOD)活性最高(P<0.05),300、600、900μg∕mL PCP添加组精液中的过氧化氢酶(CAT)活性较高(P<0.05),600μg∕mL PCP添加组精液中的谷胱甘肽过氧化物酶(GSH-Px)活性最高(P<0.05);所有添加组的精子DNA甲基化水平均显著低于对照组(P<0.05)。综上,添加适量的PCP可以改善上海白猪的精液冷冻保存效果,也能够降低冷冻后精子的DNA甲基化水平。

【Abstract】 To study the effects ofPoria cocospolysaccharides(PCP) on cryopreservation of Shanghai White Pig semen and DNA methylation of sperm,24 ejaculates were collected by artificial vagina from 8 Shanghai White Pigs. Semen samples were diluted with tris-based extender supplemented with different concentrations of PCP(0,300,600,900,1 200,1 500 μg∕mL). After thawing,sperm samples were assessed for semen quality,antioxidant activities and DNA methylation of sperm. The results showed that the addition of 600 μg∕mL PCP into the extender led to a higher semen motility in comparison to the control(P< 0. 05). The addition of 600 μg∕mL PCP into the extender led to a higher percentages of plasma membrane and acrosome integrities,and mitochondrial activity of sperm(P< 0. 05). Compared with the controls,the addition of 600 μg∕mL and 900 μg∕mL PCP significantly decreased the expressions of reactive oxygen species(ROS) and malondialdehyde(MDA) in sperm(P< 0. 05),the addition of 600 μg∕mL PCP significantly increased the super oxidase dismutase(SOD) level in sperm(P< 0. 05),the addition of 300,600 μg∕mL and 900 μg∕mL PCP significantly increased the catalase(CAT) activity in sperm(P< 0. 05),and the addition of 600 μg∕mL PCP significantly increased the activity of glutathione peroxidase(GSH-Px) in sperm(P< 0. 05). A significant decrease in the sperm DNA methylation levels was observed in all groups with the addition of PCP compared to the control(P< 0. 05). In summary,the addition of PCP to the extenders enhanced the post-thawed semen quality of Shanghai White Pig sperm and also improved the DNA methylation of sperm.
