参附汤治疗慢性阻塞性肺疾病的作用机制分析

【摘要】 目的 分析参附汤治疗慢性阻塞性肺疾病(COPD)的作用机制。方法 通过TCMSP、Genecards等数据库获取参附汤活性成分和COPD疾病靶点,通过Cytoscape软件构建“药物-活性成分-关键靶点-疾病”网络,筛选参附汤治疗COPD关键靶点;运用DAVID数据库对参附汤治疗COPD关键靶点关键靶点进基因本体论、京都基因与基因组百科全书富集分析。结果 参附汤活性成分主要有山奈酚、β-谷甾醇及豆甾醇等,COPD疾病靶点有108个。“药物-成分-关键靶点-疾病”网络中筛选出ABCB1、MMP2及CCND1等19个参附汤治疗COPD关键靶点。参附汤主要通过磷脂酰肌醇3激酶-蛋白激酶B(PI3K-AKT)信号通路、丝裂原活化蛋白激酶(MAPK)信号通路及巨细胞病毒感染等154条通路影响COPD的炎症反应、组织重塑、细胞增殖和凋亡等多种病理生理过程。结论 参附汤可能通过调节ABCB1、MMP2及CCND1表达,调控PI3K-AKT信号通路和MAPK信号通路,减轻COPD患者的肺部炎症反应。

【Abstract】 Objective To analyze the mechanism of action of Shenfu decoction in the treatment of chronic obstructive pulmonary disease(COPD). Methods The active ingredients of Shenfu decoction and the disease targets of COPD were obtained through databases such as TCMSP and Genecards. A "drug-active component-key target-disease" network was constructed using Cytoscape software to screen the key targets of Shenfu decoction in the treatment of COPD. DAVID database was used to conduct gene ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analysis on the key targets of Shenfu decoction in the treatment of COPD. Results The main active ingredients of Shenfu decoction included kaempferol, β-sitosterol, and stigmasterol, etc. There were 108 disease targets for COPD. Nineteen key targets of Shenfu decoction in the treatment of COPD were screened out from the "drug-component-key target-disease" network, including ABCB1, MMP2, and CCND1, etc. Shenfu decoction mainly affected the inflammatory response, tissue remodeling, cell proliferation and apoptosis and other pathophysiological processes of COPD through 154 pathways such as the phosphatidylinositol 3-kinase-protein kinase B(PI3K-AKT) signaling pathway, mitogen-activated protein kinase(MAPK) signaling pathway, and cytomegalovirus infection. Conclusion Shenfu decoction may alleviate the pulmonary inflammatory response in COPD patients by regulating the expression of ABCB1, MMP2 and CCND1 and modulating the PI3K-AKT signaling pathway and MAPK signaling pathway.
