超声辅助乙醇碱液法结合MCI分离柱优化陈皮中橙皮苷的提取及纯化工艺
Extraction and Purification Process of Hesperidin from Chenpi by Ultrasound Assisted Ethanol Alkali Solution Method Combined with MCI Separation Column
【摘要】 研究以陈皮为原料,开发了一种基于超声辅助乙醇碱液提取与MCI柱层析纯化相结合的橙皮苷高效制备工艺。通过正交实验优化了橙皮苷的提取工艺,确定最佳工艺条件为超声温度70℃、pH=11、提取时间150 min、水溶剂倍数为10的条件下。经减压抽滤浓缩滤液后,采用MCI分离柱,以50%甲醇溶液为洗脱剂提取橙皮苷。实验结果表明,该新工艺显著提高了橙皮苷的分离效率,最终产物提取率达到2%,具有操作简便、产物纯度高等特点,为天然产物中类黄酮化合物的工业化提取提供了创新性技术方案,在医药和功能性食品领域具有重要应用价值。
【Abstract】 This study developed an efficient preparation process for hesperidin using tangerine peel as raw material, combining ultrasound-assisted ethanol-alkaline extraction with MCI column chromatography purification. Through orthogonal experiments, the crude extraction process was optimized, with optimal conditions identified as: ultrasonic temperature 70 ℃, p H=11, extraction time 150 min, and10-fold water solvent. After vacuum filtration and concentration of the filtrate, MCI separation column was employed using 50% methanol solution as eluent for hesperidin purification. Experimental results demonstrate that this novel process significantly improves separation efficiency, achieving a final product extraction rate of 2%. Characterized by operational simplicity and high product purity, this innovative technical scheme provides an industrial extraction method for flavonoid compounds from natural products, holding significant application value in pharmaceutical and functional food industries.
