多种花椒提取物对于LL-37引起的瘙痒模型的作用及机制初探

【摘要】 目的 探究花椒提取物对于由LL-37诱导的瘙痒模型的缓解作用及其机制。方法 采用LL-37诱发小鼠瘙痒行为,分别给予花椒水提取物、花椒油提取物及2%山椒素处理,通过观察小鼠搔抓行为学变化,并进行基因表达分析,评估多种花椒提取物对于缓解瘙痒的效果及对炎症、瘙痒相关基因的影响。结果 花椒水提取物组(WEZB组)、花椒油提取物组(EOZB组)和2%山椒素组(HAS组)均显著减少了小鼠的搔抓行为,其中2%山椒素表现出最佳抗瘙痒效果。实时荧光定量PCR结果显示,WEZB组、EOZB组和HAS组中,多种炎症和瘙痒相关的mRNA水平相较于对照组显著降低;其中所有花椒提取物均可显著下调TRPV1、TRPV4、IL-31;而HAS组不仅瘙痒的相关基因表达有显著下调,炎性相关因子(TNF-α、IL-1β)的表达也有显著降低。结论 花椒提取物具有改善瘙痒行为的潜力,为抗瘙痒药物研发提供新靶点,但其临床应用需进一步验证安全性与有效性,其作用机制仍待进一步研究。

【Abstract】 Objective To investigate the ameliorative effects and mechanisms of Zanthoxylum bungeanum(Sichuan pepper) extracts on an LL-37-induced itching model.Methods Itch behavior in mice was elicited using LL-37,followed by administration of aqueous Zanthoxylum extract, Zanthoxylum oil extract, and 2% hydroxy-alpha-sanshool.Mice scratching behavior was observed, and gene expression analyses were conducted to evaluate the efficacy of various Zanthoxylum extracts in alleviating itch and their impact on genes associated with inflammation and itch.Results The groups treated with Sichuan pepper water extract, Sichuan pepper oil extract, and 2% sanshool all significantly reduced scratching behavior in mice, with 2% sanshool demonstrating the best anti-pruritic effect.Real-time quantitative PCR results showed that multiple mRNAs associated with inflammation and pruritus were significantly downregulated in the Sichuan pepper water extract group, Sichuan pepper oil extract group, and 2% sanshool group compared to the control group.Notably, all Sichuan pepper extracts significantly downregulated TRPV1,TRPV4,and IL-31 mRNA levels.Moreover, in addition to significant reduction of the expression of pruritus-related genes, the 2% sanshool group also markedly decreased the expression of inflammatory-related factors(TNF-α,IL-1β).Conclusion Extracts from Sichuan pepper have a potential for improving pruritic behavior, providing new targets for the development of anti-pruritic drugs.However, further validation of safety and efficacy for clinical application is required, and the mechanisms of action warrant additional investigation.
