鱼类水解蛋白肽对铜胁迫后大口黑鲈抗氧化能力及脂肪合成酶活性的影响
Effects of Fish Hydrolyzed Protein Peptides on Antioxidant Capacity and Lipid Synthetase Activity of Largemouth Bass(Micropterus salmoides) Exposed to Copper Stress
【摘要】 为探讨鱼水解蛋白肽(HFP)缓解铜胁迫对大口黑鲈(Micropterus salmoides)抗氧化能力及脂肪合成酶活性的影响,在水温(24.0±1.0)℃下,将体质量(9.51±0.01)g的大口黑鲈饲养在0.6 m×0.6 m×0.8 m的循环水养殖缸内,投喂以鱼粉、鸡肉粉、豆粕、大豆浓缩蛋白和棉籽蛋白为蛋白源,以鱼油和豆油为脂肪源配制的基础饲料,分别添加0 g/kg(CK)、10 g/kg(HFP1)、30 g/kg(HFP3)和50 g/kg HFP(HFP5)。饲养56 d后,在水中投放0.25 mg/L Cu(纯度99%的硫酸铜配制)14 d,关闭循环水系统。铜胁迫结束时测定大口黑鲈肠道、胃和幽门盲囊的抗氧化酶、脂肪合成酶和ATP酶活性。结果表明,与CK组相比,添加不同水平HFP组大口黑鲈肠道的总抗氧化能力(T-AOC)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活力和丙二醛(MDA)含量显著降低(P<0.05);HFP3组和HFP5组大口黑鲈胃CAT活力和MDA含量显著降低(P<0.05);添加不同水平HFP显著降低大口黑鲈幽门盲囊T-AOC和CAT活性(P<0.05)。添加不同水平HFP显著降低大口黑鲈肠道苹果酸酶(ME)和6-磷酸葡萄糖酸脱氢酶(6PGD)活性(P<0.05),HFP5组大口黑鲈肠道脂肪酸合成酶(FAS)和葡萄糖6-磷酸脱氢酶(G6PD)活性显著低于CK组(P<0.05);添加不同水平HFP显著降低大口黑鲈胃ME和G6PD活性(P<0.05),HFP3组和HFP5组大口黑鲈胃FAS和6PGD活性显著低于CK组(P<0.05);HFP5组大口黑鲈幽门盲囊FAS和G6PD活性显著低于CK组(P<0.05);HFP1组、HFP3组和HFP5组大口黑鲈肠道、胃和幽门盲囊脂肪酶(LPS)活性显著升高(P<0.05);HFP3组和HFP5组大口黑鲈胃和幽门盲囊ATP酶水平显著高于CK组(P<0.05)。综上所述,HFP可通过提高铜胁迫后大口黑鲈肠道、胃和幽门盲囊的抗氧化酶能力,改善脂肪合成相关酶活性和ATP酶水平,减轻大口黑鲈抗氧化应激损伤,促进大口黑鲈在铜胁迫下内环境稳态的恢复。
【Abstract】 In order to investigate the effects of fish protein hydrolyzed peptides(HFP) on the antioxidant capacity and lipid synthetase activity of largemouth bass(Micropterus salmoides) alleviating copper stress, largemouth bass with body weight of(9.51 ±0.01) g were reared in a recirculating aquaculture tank of 0.6 m ×0.6 m × 0.8 m at water temperature of(24.0 ±1.0) ℃, and fed basic isonitrogenous and isoenergetic diet prepared by soybean protein concentrate and cottonseed protein as protein sources, and fish oil and soybean oil as fat sources, and supplemented with 0 g/kg(CK), 10 g/kg(HFP1), 30 g/kg(HFP3) and 50 g/kg HFP(HFP5) for 56 d.Then the largemouth bass was exposed to 0.25 mg/L Cu(prepared with copper sulfate of 99% purity) for 14 d, without the circulating water. At the end of copper stress, the activities of antioxidant enzymes, lip synthase, and ATPase were measured in the intestine,stomach, and pyloric caeca of largemouth bass. The results showed that the largemouth bass fed the diets supplemented with different levels of HFP had significant in total antioxidant capacity(T-AOC), catalase(CAT), glutathione peroxidase(GSH-Px) activities, and malondialdehyde(MDA) content in the intestine compared with the CK group(P < 0.05). There were significant decreases in CAT activity and MDA content in the stomach of largemouth bass in the HFP3 and HFP5 groups, and significant decreases in T-AOC and CAT activities in the pyloric caeca of largemouth bass(P < 0.05). Supplementation with different levels of HFP led to significantly decreases in the intestinal malic enzyme(ME) and 6-phosphogluconate dehydrogenase(6PGD) activities(P < 0.05), with significantly lower intestinal fatty acid synthase(FAS) and G6PD activities of largemouth bass in the HFP5 group than those in the CK group(P <0.05). There were significant decreases in the activities of ME and glucose-6-phosphate dehydro genase G6PD in the stomach of largemouth bass(P < 0.05), with significantly lower activities of FAS and 6PGD in the stomach of largemouth bass in the HFP3 and HFP5 groups than those in the CK group(P < 0.05). The activities of FAS and G6PD in the pyloric caeca were found to be significantly lower in HFP5 group than those in the CK group(P < 0.05). The LPS activity in the intestine, stomach and pyloric caeca was significantly increased in the HFP1, HFP3 and HFP5 groups(P < 0.05), and the ATPase level in the stomach and pyloric caeca was significantly higher in the HFP3 and HFP5 groups than those in the CK group(P < 0.05). In conclusion, HFP led to improve the antioxidant enzyme activity and ATPase level in the intestine, stomach, and pyloric caeca, to alleviate antioxidant stress damage and to promote the restoration of homeostasis of largemouth bass under copper stress.
