n-3多不饱和脂肪酸对驯养的异齿裂腹鱼繁殖功能障碍的影响
Effects of n-3 Polyunsaturated Fatty Acids on Reproductive Dysfunction in Domestic Schizothorax o′connori
【摘要】 异齿裂腹鱼(Schizothorax o′connori)是青藏高原雅鲁藏布江特有鱼类,具有重要的经济价值和生态价值。本研究采用酶联免疫和HE染色的方法,检测VI期雌性驯养和野生异齿裂腹鱼血清中性激素水平并比较前肠组织的形态差异;通过气相色谱分析的方法,检测VI期雌性驯养和野生异齿裂腹鱼肝脏和卵母细胞中脂肪酸的组成及含量。结果显示:驯养异齿裂腹鱼雌激素(E)含量极显著低于野生异齿裂腹鱼(P<0.01);孕酮(P)含量显著高于野生异齿裂腹鱼(P<0.05);睾酮(T)含量显著低于野生异齿裂腹鱼(P<0.05)。在肝脏和卵母细胞中,驯养异齿裂腹鱼α-亚麻酸(ALA)含量均显著高于野生异齿裂腹鱼(P<0.05);而二十碳五烯酸(EPA)含量均极显著低于野生异齿裂腹鱼(P<0.01);二十二碳六烯酸(DHA)含量差异不显著,但均表现出驯养低于野生的趋势。综上所述,VI期驯养异齿裂腹鱼血清中E和T水平较低,表现出明显的繁殖功能障碍,且肝脏和卵母细胞中n-3 PUFAs的组成及含量是造成其繁殖功能障碍的重要原因之一。
【Abstract】 Schizothorax o′ connori is an endemic fish of the Brahmaputra River on the Qinghai-Tibet Plateau, and has important economic and ecological value. This study utilized ELISA and HE staining techniques to assess serum sex hormone levels and foregut morphology in stage VI domestic and wild S. o′connori. Additionally, gas chromatography was employed to analyze fatty acid profiles in the liver and oocytes of both domestic and wild variants. The results revealed distinctive disparities between domestic and wild specimens. Domestic S. o′ connori exhibited notably lower estrogen levels than their wild counterparts(P < 0.01). Conversely,progesterone levels were significantly higher(P < 0.05), while testosterone levels were markedly lower than those in the wild population(P < 0.05). Further scrutiny of fatty acid content unveiled significant differences. Domestic specimens displayed significantly higher ALA content(P < 0.05) but lower EPA levels(P < 0.01) in both liver and oocytes compared to their wild counterparts. While no substantial variance in DHA content was observed(P > 0.05), domesticated S. o′connori showed comparatively lower DHA levels. In summary, the reduced levels of estrogen and testosterone in the serum of domestic S. o′connori strongly indicate evident reproductive dysfunction. Moreover, the altered composition and decreased content of n-3 PUFAs in the liver and oocytes appear to be significant contributing factors to this reproductive impairment.
