亲水相互作用色谱-串联质谱法测定7种乳粉中17种氨基酸

Determination of 17 amino acids in seven types of milk powder using hydrophilic interaction high-performance liquid chromatography-tandem mass spectrometry

【摘要】 文章使用6 mol/L盐酸将乳粉中蛋白质水解为游离氨基酸,XBridge BEH Amide色谱柱对目标物进行分离,采用四级杆串联线性离子阱质谱仪基于正离子电离模式、多反应监测数据采集模式、峰面积外标法定量模式对目标物进行检测。结果表明,乳粉样品在稀释1×10~5倍情况下,优化后的色谱方法可将17种氨基酸有效分离,目标物的基质效应基本被消除,在0.5~8μmol/L浓度范围内线性关系良好,检出限和定量限分别为0.02~0.99 g/kg和0.08~3.27 g/kg。使用该方法成功分析了市售牦牛乳粉、山羊乳粉、绵羊乳粉、骆驼乳粉、马乳粉、驴乳粉和牛乳粉中17种氨基酸的含量,检测结果与国家标准方法相比无统计学差异。所建方法具有操作简单、灵敏度高、结果准确等技术优势,适用于分析乳粉中氨基酸组成。

【Abstract】 The protein in milk powder was hydrolyzed to free amino acids using 6 mol/L hydrochloric acid, and the target compounds were subsequently separated using an XBridge BEH Amide chromatographic column. Quantitative analysis of these amino acids was performed using a quadrupole tandem linear ion trap mass spectrometer under positive ion multiple reaction monitoring acquisition mode,using the peak area external standard method for quantification. The optimized chromatographic method effectively separated the 17 amino acids in milk powder samples diluted 10~5 times,effectively eliminating matrix effects.A strong linear relationship was observed within the concentration range of 0.5-8.0 μmol/L. The limits of detection and limits of quantification ranged from 0.02-0.99 g/kg and 0.08-3.27 g/kg,respectively. This method was applied to analyze the amino acid contents in various commercial milk powders,including yak,goat,sheep,camel,horse,donkey,and cattle milk powders.The results were statistically comparable to those obtained using the national standard method.The established technique offers simplicity in operation,high sensitivity,and accurate results,making it well-suited for analyzing the amino acid composition in milk powder.
