槲皮素缓解神经性耳鸣的作用及机制
Role and mechanism of quercetin in alleviating neurogenic tinnitus
【摘要】 目的 分析槲皮素缓解神经性耳鸣的作用及机制。方法 选取C57BL/6小鼠20只,按随机数字表法分为预处理组、处理组、模型组、正常对照组,各5只。预处理组、处理组及模型组构建神经性耳鸣小鼠模型,正常对照组不建模。预处理组小鼠在建模前2 h给予槲皮素25 mg/kg灌胃,建模后第2天开始,预处理组和处理组小鼠每天给予槲皮素25 mg/kg灌胃,模型组和正常对照组小鼠给予等体积生理盐水灌胃,均持续1周。比较4组小鼠听觉[听性脑干反应(ABR)阈值]、耳鸣行为学[间歇前抑制惊跳反射抑制率(GPIAS%)],处死小鼠后采集耳蜗组织,检测组织中炎性因子[肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)]mRNA表达和蛋白表达水平。结果 与正常对照组比较,模型组小鼠在4、6、8 kHz刺激下ABR阈值升高,而预处理组和处理组低于模型组,且预处理组低于处理组(P<0.05)。与正常对照组比较,模型组GPIAS%降低,而预处理组和处理组高于模型组,且预处理组高于处理组(P<0.05)。与正常对照组比较,模型组小鼠TNF-α和IL-1β mRNA表达和蛋白表达水平均升高,而预处理组和处理组低于模型组,且预处理组低于处理组(P<0.05)。结论 槲皮素能改善神经性耳鸣小鼠的听力和耳鸣行为,其作用机制可能与槲皮素抑制神经性耳鸣小鼠炎性反应有关。
【Abstract】 Objective To analyze the role and mechanism of quercetin in alleviating neurogenic tinnitus. Methods Twenty C57BL/6 mice were selected and randomly divided into pretreatment group, treatment group, model group and normal control group, with 5 mice in each group. The pretreatment group, treatment group and model group were used to establish an animal model of neurogenic tinnitus, while the normal control group was not modeled. The pretreatment group received quercetin(25 mg/kg) via gavage 2 hours before model establishment. Starting from the second day post-modeling, the pretreatment group and treatment group were administered quercetin(25 mg/kg)daily via gavage, while the model group and normal control group received an equivalent volume of saline via gavage, all treatments were maintained for 1 week. The auditory function(ABR)and tinnitus behavior(GPIAS%)of the four groups were compared. After euthanizing the mice, cochlear tissues were collected to measure the mRNA and protein expression levels of inflammatory factors(TNF-α and IL-1β) in the tissues.Results Compared with the normal control group, the ABR thresholds of mice in model group was elevated at 4, 6 and 8 kHz stimuli, while, the pretreatment group and treatment group were lower than the model group, with the pretreatment group was lower than the treatment group(P<0.05). Compared with the normal control group, GPIAS% in the model group showed reduced, nevertheless, the pretreatment group and treatment group were higher than the model group, with the pretreatment group was higher than the treatment group(P<0.05). Compared with the normal control group, mRNA and protein expression levels of TNF-α and IL-1β were upregulated in model group, the pretreatment group and treatment group were lower than the model group, wheresa the pretreatment group was lower than the treatment group(P<0.05). Conclusion Quercetin effectively improves auditory function and mitigates tinnitus-related behaviors in mice neurogenic tinnitus hearing loss, the underlying mechanism may involve its anti-inflammatory properties, as evidenced by the suppression of TNF-α and IL-1β expression.
