枸杞多糖对脂多糖诱导BV2细胞帕金森病炎症模型的作用及其机制研究

【摘要】 目的 观察枸杞多糖(LBP)对脂多糖(LPS)诱导的BV2细胞系(BV2细胞)帕金森病炎症模型的影响及相关机制。方法 将BV2细胞分为正常组、LPS组和不同浓度的LBP组。采用不同浓度的LBP(15μg/mL、30μg/mL和60μg/mL)进行实验,LPS组使用1μg/mL的LPS刺激BV2细胞,2 h后再加入不同浓度的LBP,共孵育24 h;采用CCK-8检测各组细胞的活性;采用ELISA法检测各组BV2细胞上清液中TNF-α和IL-1β含量;采用Western blot法检测各组p65、p-p65及ERK1/2表达。结果 CCK-8实验检测发现,不同LPS干预浓度对BV2细胞无毒性;与正常组比较,LPS组BV2细胞上清液中TNF-α和IL-1β蛋白含量显著升高(P<0.05);与LPS组比较,LBP组BV2细胞上清液中TNF-α和IL-1β蛋白含量显著降低(P<0.05)。与正常组比较,LPS组BV2细胞p65、p-p65及ERK1/2蛋白表达显著升高(P<0.05);与LPS组比较,LBP组BV2细胞p65、p-p65及ERK1/2蛋白表达显著下降(P<0.05)。结论 LBP可能减轻LPS诱导的BV2细胞损伤,其机制可能与下调p65、p-p65及ERK1/2蛋白相关,60μg/mL可能为LBP最佳的干预浓度。

【Abstract】 Objective To observe the effect of lycium barbarum polysaccharides(LBP) on lipopolysaccharide(LPS) induced BV2 cell line and its mechanism.Methods BV2 cells were divided into normal group, LPS Group and LBP Group. Different concentrations of LBP(15 μg/mL,30 μg/mL and 60 μg/mL) were used in the experiment. The BV2 cells were stimulated with 1 μg/mL LPS and then added with different concentrations of LBP. After incubation for 24 hours, CCK-8 was used to detect the activity of BV2 cells, and ELISA was used to detect the contents of TNF-α and Il-1β in the supernatant of BV2 cells. The expression of p65,p-p65 and ERK1/2 were detected by wtslern blot.Results CCK8 assay showed that different intervention concentrations of LPS were non-toxic to BV2 cells.Compared with normal group, the protein contents of TNF-α and Il-1β in the supernatant of BV2 cells in LPS group were significantly increased(P<0.05);the protein contents of TNF-α and Il-1β in the supernatant of BV2 cells in LBP group were significantly lower than those in the control group(P<0.05);The protein expressions of p65,p-p65 and ERK1/2 in BV2 cells in LPS group were significantly higher than those in the control group(P<0.05) Compared with LPS Group, the expression of p65,p-p65 and ERK1/2 protein in BV2 cells in LBP group were significantly decreased(P<0.05).Conclusion LBP can regulate LPS-induced BV2 cell injury and inhibit inflammatory reaction, which may be related to down regulation of p65,p-p65 and ERK1/2 signaling pathway. Low concentration of LBP has little effect on BV2 cell and 60 μg/mL LBP is the best concentration.
